Human induced pluripotent stem (iPS) cells can be passaged as single cells in the Cellartis DEF-CS Culture System. A single GFP-actin iPS cell was isolated and placed in the well of a culture dish. Twenty-four hours after seeding, morphology was assessed by fluorescence microscopy at 20x (Panel A) and 40x (Panel B) magnification. Sixteen days later, the single GFP-actin iPS cell had proliferated into numerous cells as evidenced by microscopic observation at 4x (Panel C), 10x (Panel D), 20x (Panel E), and 40x (Panel F) magnification.

Immunocytochemistry analysis of hepatocyte differentiation. hiPS cells were differentiated into functional hepatocytes using the Cellartis iPS Cell to Hepatocyte Differentiation System. Hepatocytes were immunostained to detect early hepatic markers HNF4α (red, nuclear) and CK18 (green) on days 14 and 21. As the hepatocytes matured, expression of liver-specific markers CYP3A (red, cytoplasmic) and Albumin (green) increased as seen on day 28, as expected. Cell nuclei were stained with DAPI (blue).

Human Mitochondrial DNA (mtDNA) Monitoring Primer Set is designed to quantify the relative number of copies of human mitochondrial DNA (mtDNA) by real-time PCR, using nuclear DNA (nDNA) content as a standard. This set contains primer pairs for the amplification of four regions: two primer pairs for the detection of mtDNA and two primer pairs for the detection of nDNA. Each primer is specifically designed and will not amplify pseudogenes. As the primers are human-specific and will not amplify DNA from mice, human mtDNA content can be monitored in iPS cells or other cells cultured with feeder cells of mouse origin.
