Magnetic bead-based isolation of genomic DNA from tissue—NucleoMag Tissue
NucleoMag Tissue is designed for manual or automated small-scale preparation of genomic DNA from human and animal tissue, cultured cells, or bacteria, in a 96-well format. After lysis of the sample, genomic DNA is selectively bound to NucleoMag Beads. Contaminants are washed away and the DNA can be eluted from the beads under low-salt conditions.
NucleoMag Tissue is designed for manual or automated small-scale preparation of genomic DNA from human and animal tissue, cultured cells, or bacteria, in a 96-well format. After lysis of the sample, genomic DNA is selectively bound to NucleoMag Beads. Contaminants are washed away and the DNA can be eluted from the beads under low-salt conditions. Since the magnetic bead technology provides a closed system (no transfer of sample from one well to another during the preparation), the risk for cross contamination is reduced.
Typically, 96 samples can be purified in less than 120 minutes on common automation platforms. The kit provides reagents for the purification of 10–20 µg of genomic DNA from samples (e.g., 20 mg of tissue or 107 cells). The purified DNA is ready to use for PCR, Southern blotting, and any enzymatic reaction.

Detection of influenza A virus subtype H1N1
In this study, we describe the automated use of the NucleoMag Virus kit on the KingFisher Flex magnetic separator for the isolation of viral nucleic acids and subsequent detection by PCR. For kit evaluation, A/H1N1 samples from an interlaboratory test, as well as clinical samples, were used for kit evaluation. The data demonstrates the excellent quality of the eluted viral RNA which is suitable for downstream RT-PCR.
