Magnetic bead-based isolation of viral RNA/DNA and bacterial DNA from veterinary samples—NucleoMag VET

Magnetic bead-based isolation of microbial RNA/DNA from veterinary samples.

NucleoMag VET enables manual or automated purification of viral RNA and DNA and bacterial DNA using proven NucleoMag magnetic bead technology. The kit is designed for common veterinary sample material, such as animal whole blood, serum/plasma, feces, ear notches, tissue, or swabs. After a sample-specific pre-lysis procedure, microbial nucleic acids are purified from 200 µl of liquid or homogenized sample. The entire NucleoMag VET protocol can be automated on common liquid handling platforms.

NucleoMag VET enables manual or automated purification of viral RNA and DNA and bacterial DNA using proven NucleoMag magnetic bead technology. The kit is designed for common veterinary sample material, such as animal whole blood, serum/plasma, feces, ear notches, tissue, or swabs. After a sample-specific pre-lysis procedure, microbial nucleic acids are purified from 200 µl of liquid or homogenized sample. The entire NucleoMag VET protocol can be automated on common liquid handling platforms.

The NucleoMag principle is similar to silica membrane technology: nucleic acids are bound to magnetic beads under chaotropic salt conditions, contaminants are washed away with ethanolic wash buffers, and pure RNA/DNA are eluted from the beads using elution buffer. NucleoMag technology can be used either with static-pin separators (e.g., NucleoMag SEP) or with magnetic separators integrated into robotic workstations.

NucleoMag VET prefilled plates are prefilled 96 deep-well plates specifically for use on the IsoPure Mini system for a simplified automation workflow for 8–32 veterinary samples in parallel. Binding Buffer VEB, NucleoMag B-Beads, Wash Buffer VEW1, VEW2, 80% ethanol, and elution buffer VEL are prefilled into their respective wells. Sample preparation/lysis is performed according to the standard procedure of the NucleoMag VET kit. Subsequently, lysed samples are transferred to the reagent wells containing binding buffer VEB and NucleoMag B-Beads for further processing on the IsoPure Mini instruments. The purified RNA/DNA, eluted in 100 µL VEL, can be used directly as a template for RT-PCR, PCR, or any enzymatic reaction.

NucleoMag VET is designed for the isolation of viral DNA and RNA and bacterial DNA from 100–200 µl inputs of veterinary samples, such as serum or plasma, whole blood, homogenized tissue suspensions, etc. The kit procedure is based on the reversible adsorption of nucleic acids to paramagnetic beads under appropriate buffer conditions.

Sample lysis is achieved by incubation with Lysis Buffer VL1 containing chaotropic ions in combination with Proteinase K digestion. Binding of nucleic acids to paramagnetic beads then occurs when NucleoMag B-Beads and Binding Buffer VEB are added to the lysate. After magnetic separation, several wash steps are performed to remove contaminants and salts, followed by air-drying and elution of nucleic acids under low-salt conditions, yielding DNA/RNA that is ready for direct use in downstream applications. The NucleoMag VET protocol can be performed manually or automated on common liquid handling instruments.
