Magnetic bead-based isolation of viral RNA/DNA and bacterial DNA—NucleoMag Pathogen

The NucleoMag Pathogen kit enables purification of viral RNA and DNA and microbial DNA using magnetic beads. The kit is designed for common clinical samples, such as whole blood, serum or plasma, feces, tissue, or swabs. After a sample-specific pre-lysis procedure, 200 µl of liquid or homogenized sample is used in the purification procedure.

The NucleoMag product family is based on magnetic bead technology. Nucleic acids are bound to the magnetic beads under chaotropic salt conditions, contaminants are washed away with ethanolic wash buffers, and pure RNA and DNA can be eluted from the beads using elution buffer.

The NucleoMag technology can easily be adapted to common liquid handling instruments. It can be used either with static pin separators (e.g., NucleoMag SEP) or magnetic separators integrated into robotic workstations.

Dilution series of 10^-3–10^-4 of feces samples were prepared for sensitivity screening using three replicates. Nucleic acid extraction was performed with the NucleoMag Pathogen kit and the competitor kit "R". PCR analysis was performed using the RIDA GENE Viral Stool Panel I (R-Biopharm) and RealStar Clostridium difficile PCR Kit 1.0 (Altona Diagnostics). Back Sensitivity screening for Hepatitis B virus (HBV) and Hepatitis C virus (HCV) Sensitivity screening for Hepatitis B virus (HBV) and Hepatitis C virus (HCV). Dilution series (1:10) of human plasma (200 �l, with original virus titer as shown) were extracted in triplicate with the NucleoMag Pathogen kit and tested with the RealStar HBV PCR Kit 1.0 and the HCV RT-PCR Kit 1.0 (Altona Diagnostics). The NucleoMag Pathogen kit demonstrates a sufficient sensitivity for Hepatitis B virus (HBV) and Hepatitis C virus (HCV) detection in human plasma sample material. The internal control was extracted and recovered as expected. PCR inhibitors were not detected. Back Reliable detection of viral RNA Reliable detection of viral RNA. A Norovirus-positive stool sample and various dilutions of the sample were processed for viral RNA purification using the NucleoMag Pathogen kit and competitor kit "R". Purified viral RNA was analyzed by qPCR. Back Superior detection of SARS-CoV-2 from human swab samples Viral RNA from 19 confirmed SARS-CoV-2-positive swab samples was extracted in duplicate using NucleoMag Pathogen and a competitor kit (Q). Viral RNA was quantified using a SARS-CoV-2-specific qRT-PCR assay (qScript™ XLT One-Step RT-qPCR ToughMix® + nCoV IP4 assay; Institute Pasteur, Paris). For each sample tested, the resulting CT values indicated that the NucleoMag Pathogen kit performed as well as (3/19 samples), or better than the competitor kit (16/19 samples). For two samples tested, detection of SARS-CoV-2 RNA was possible with NucleoMag Pathogen but not with the competitor kit. Back Consistent qRT-PCR amplification of SARS-CoV-2 RNA extracted from different sample types using NucleoMag Pathogen Dilution series were prepared for three different sample types (nasal swab, oral swab, saliva) using inactivated SARS-CoV-2 virus. Viral RNA was extracted using NucleoMag Pathogen on a KingFisher Flex system and the RNA was quantified by qRT-PCR (AgPath-ID™ One-Step RT-PCR mix + nCoV IP4 assay, Institute Pasteur, Paris). For each sample type, viral RNA was detected consistently and reliably over a 5-log range of dilutions. Back Superior detection of SARS-CoV-2 from human saliva samples Viral RNA was extracted from 200-μl inputs of diluted SARS-CoV-2-positive saliva samples in duplicate using NucleoMag Pathogen (MN) and the MagMAX™ Viral RNA Isolation Kit (T), respectively, and quantified by qRT-PCR.
