A fast way to purify plasmids for transfection—NucleoSnap Plasmid Midi
Endotoxins are copurified during plasmid preparations from bacterial lysates. Since they interfere with eukaryotic cell survival, endotoxin reduction is essential prior to cell transfection. Macherey-Nagel has developed a new snap-off column format and a novel buffer chemistry to enable vacuum-processed isolation of transfection-grade plasmid DNA in a midiprep format. See our NucleoSnap Plasmid Midi tech note showing that you can purify 250 μg of plasmid DNA in only 35 minutes.
- New column design (snap-off column) for vacuum processing of large sample volumes
- Transfection-grade plasmid DNA for sensitive downstream applications
- Isolate 250 μg of plasmid DNA in only 35 minutes
|Processing||Vacuum processing (e.g., using the NucleoVac 24 Vacuum Manifold), or elution by centrifugation|
|Endotoxin content||<50 EU/µg DNA (EU = Endotoxin Units)|
|Starting material||Up to 50 ml E. coli culture (A600 = 5)|
|Lysate clarification||NucleoSpin Plasmid Filter Columns (2 min centrifugation)|
|Vector Size||≤25 kb|
|Typical yield||250 µg (50 ml culture, A600 = 4, high-copy plasmid)|
|Elution volume||200–500 µl|
|Preparation time||35 min/6 preps|
|Binding capacity||1.5 mg|
- Typical downstream applications: transfection of common cell lines (lipofection, electroporation)
- Also suitable for standard molecular biology applications such as cloning and sequencing

Isolate up to 700 µg of transfection-grade plasmid DNA in only 35 minutes
Using novel chemistry and a unique snap-off column design, NucleoSnap Plasmid Midi provides rapid purification of up to 700 µg of transfection-grade plasmid. The snap-off columns allow processing of lysate from large 50-ml culture volumes, and multiple samples can be rapidly processed by vacuum using our NucleoVac 24 Vacuum Manifold.Learn more Other plasmid kits
