The protocol begins with mechanical disruption of your plant/fungal samples (using MN Bead Tubes or Bead Plates, grinding in liquid nitrogen, or any other suitable disruption method) in lysis buffer containing large amounts of chaotropic ions to effectively disrupt tough plant/fungal material. This lysis buffer also immediately inactivates RNases, which are present in virtually all biological materials. The NucleoSpin 96 Plant and Fungi Filter Plate quickly and efficiently removes plant/fungal debris, and a binding solution is added, which creates appropriate binding conditions that favor the adsorption of intact RNA to the silica membrane. Washing steps with two different specialized buffers remove salts, metabolites, and macromolecular cellular components. The resulting pure, high-quality RNA is eluted from the silica membrane, ready for downstream transcriptomic analysis.
