Cleanup and concentration of large genomic DNA fragments
The NucleoSpin gDNA Clean-up and NucleoSpin gDNA Clean-up XS kits provide the easiest and fastest way to clean up and concentrate large genomic DNA fragments from enzymatic reactions or prepurified samples (extracted using organic solvents such as phenol/chloroform), or to perform buffer exchange.
These kits can be used to remove contaminants such as salts, enzymes, nucleotides, labeling reagents, etc., by selective binding to a silica membrane in a spin-column format using a simple bind-wash-elute procedure. The proven silica-membrane technology is an effective alternative to the widely used Centricon concentrator columns for cleaning and concentrating samples extracted by standard "organic" methods.
NucleoSpin gDNA Clean-up includes mini spin columns (50–100 µl elution volume) to purify standard samples (<150 µl aqueous DNA solution containing <25 µg DNA), while NucleoSpin gDNA Clean-up XS includes XS columns (6–10 µl elution volume) to purify very small samples (<400 µl aqueous DNA solution containing <2 µg DNA). The NucleoSpin gDNA Clean-up XS Column contains a unique silica membrane that allows the smallest elution volumes. Due to its high sensitivity, the XS kit is particularly well-suited for purifying trace levels of DNA from forensic samples. These kits reduce sample preparation time to 15–20 minutes, compared to >1 hour for ultrafiltration devices. The purified DNA is suitable for a variety of downstream applications, including PCR, enzymatic reactions, Southern blotting, qPCR, and STR amplification.
Overview
- Efficient, effective cleanup and concentration of large DNA fragments for downstream applications
- Easier and faster sample concentration than microdialysis filtration units
- Two kit options: mini columns for standard samples and XS columns for very small (e.g., forensic) samples
- Get high recovery rates (up to 90%) with NucleoSpin gDNA Clean-up
- Get highly concentrated DNA (elute in 6–10 µl) with NucleoSpin gDNA Clean-up XS

DNA cleanup: two applications in one kit
The NucleoSpin Gel and PCR Clean-Up procedure is the easiest way to purify DNA fragments from agarose gels or PCR reactions. The kit includes one buffer for both applications, which contains a pH indicator that displays the correct pH for optimal kit performance. The purification procedure for samples from enzymatic reactions (e.g., PCR) allows fast and easy removal of enzymes, nucleotides, salts, and other impurities.
