Adenovirus (CAR) receptor booster

CAR receptor boosters

The CAR Receptor Booster renders cells that cannot usually be transduced with adenovirus susceptible to infection. It can also be used to increase adenoviral transduction efficiency in all cell types. The receptor-coated cell-derived nanovesicles combine with the plasma membrane of your target cells to temporarily elevate the density of the adenovirus receptor protein CAR on the cell surface. 

The CAR Receptor Booster renders cells that cannot usually be transduced with adenovirus susceptible to infection. It can also be used to increase adenoviral transduction efficiency in all cell types. The receptor-coated cell-derived nanovesicles combine with the plasma membrane of your target cells to temporarily elevate the density of the adenovirus receptor protein CAR on the cell surface. For example, hematopoietic cells or CHO cells, which are usually resistant to adenoviral infection, can be transduced following a simple pretreatment with CAR Receptor Booster (see data images by expanding the Details arrow in the product table below).

How does it work?

The CAR Receptor Booster consists of a vial of concentrated cell-derived nanovesicles that are densely coated with the coxsackie virus and adenovirus receptor protein (CAR). When the booster vesicles are applied to your cells (10–20 µl per well of a 6-well plate for ~2 hr), they fuse with the plasma membrane, resulting in an increased level of the receptor on the cell surface. Your target cells can then be more efficiently transduced with recombinant adenovirus.

The CAR Receptor Booster increases the transduction efficiency of target cells to infection by adenovirus. The CAR Receptor Booster consists of exosome-like particles coated with adenoviral receptor protein CAR. When applied to target cells, fusion of the particles with the host cell plasma membrane temporarily increases surface levels of CAR to allow for efficient adenoviral transduction. Additionally, CAR Receptor Booster renders cells that cannot usually be transduced with adenovirus (e.g. hematopoietic cells) susceptible to infection.

Human mesenchymal stem cells (hMSCs) that are normally resistant to transduction by adenovirus vectors are rendered susceptible by treatment with the CAR Receptor Booster. hMSCs were plated in 6 well plates 24 hr prior to transduction. CAR Receptor Booster (20 µl) was added, and the cells were incubated for 2 hr at 37°C followed by addition of Adeno-X ZsGreen1 virus (MOI~190). Cells were assayed for fluorescence activity 24-hr post transduction.

Adenoviral transduction of CHO-K1 cells is facilitated by pre-treatment with CAR Receptor Booster

Adenoviral transduction of CHO-K1 cells is facilitated by pre-treatment with CAR Receptor Booster. CHO-K1 cells were plated 24 hr prior to transduction and treated for 2 hr with 10 µl of CAR Receptor Booster according to the protocol. The cells were then incubated with Adeno-X LacZ virus (MOI=34) for 72 hr and stained for beta galactosidase expression. Only the CHO-K1 cells that were pre-treated with the CAR Receptor Booster were successfully transduced with adenovirus.

CAR Receptor Boosters are concentrated exosome-like vesicles that are applied to your target cells prior to infection with adenovirus. Boosters result in increased transduction efficiency because treatment increases the cell-surface density of the receptor recognized by the adenovirus. Using this technology, you can now even transduce hematopoietic cell types with adenovirus by simply pre-treating the cells with the CAR Receptor Booster, or you can use the technology to equalize the infection efficiency across multiple target cell types.

Overview

  • Transduce any cell type with recombinant adenovirus, including hematopoietic cells and CHO cells which are known for having a low density of the CAR receptor
  • Increase efficiency of transduction by temporarily increasing the concentration of the principal adenoviral receptor proteins on your target cells
  • Simply preincubate your target cells with these concentrated exosome-like vesicles for 2 hr prior to infection with virus
