The purity of AAV particles is an important factor for obtaining efficient and stable gene transfer in cultured cells and individual animals. However, many currently available AAV purification approaches, such as cesium chloride density gradient centrifugation and iodixanol ultracentrifugation, are time-consuming and require specialized equipment.
Our AAVpro Purification Kit Midi and Maxi (All Serotypes) (Cat. # 6675 and 6666, respectively) can be used to easily purify AAV particles from AAV-producing cell lysates through a simple precipitation and filtration protocol in about four hours.
Alternatively, AAVpro Cell & Sup. Purification Kit Maxi (All Serotypes) (Cat. # 6676) allows the researcher to purify virus particles from both AAV-producing cell lysates and culture supernatant in about seven hours. This provides higher virus yields, especially for serotypes that localize predominantly in supernatants or in both cell lysates and supernatants.

Please note that the purification method largely depends on the serotype, as different capsid types behave differently within producer cells. This affects where the virus accumulates and how it can be most effectively recovered. See the table below for examples of cellular localization of typical serotypes.
