Adenovirus purification kits

Purify adenovirus quickly and easily with our adenovirus purification kits! These kits are chromatography-based systems that enable you to purify amplified adenovirus directly from the cell pellet in less than 1.5 hours. You simply pellet the cells when the cytopathic effect (CPE) is nearly complete, lyse the cells, and purify the virus.

Our Adeno-X Maxi Purification Kit has a wide range of viral capacities, allowing you to scale up or down without difficulty.

Purify adenovirus quickly and easily with our adenovirus purification kits! These kits are chromatography-based systems that enable you to purify amplified adenovirus directly from the cell pellet in less than 1.5 hours. You simply pellet the cells when the cytopathic effect (CPE) is nearly complete, lyse the cells, and purify the virus.

Our Adeno-X Maxi Purification Kit has a wide range of viral capacities, allowing you to scale up or down without difficulty. The kit delivers as many as 1 x 1012 adenoviral particles and provides all of the buffers and materials necessary for purification. This same technology is also available in our convenient Adeno-X Mega Purification Kits with which you can produce and purify up to 1013 highly infectious viral particles (up to 3 x 1011 IFU).

Simple chromatographic procedure

Our protocol contains no ultracentrifugation steps. Instead, adenovirus is purified chromatographically using a unique membrane that selectively binds adenoviral particles. The membrane is housed in a small, single-use cartridge that fits securely on standard size Luer-Lok syringes. For added convenience, the syringe-filter assembly comes completely preassembled and ready to use. The adenovirus-containing medium is brought into the syringe through tubing attached to a one-way valve and is pushed out through the purification cartridge. As virus-containing medium passes through the purification cartridge, the adenoviral particles adhere to the membrane and are effectively removed from the solution. The bound particles are then washed and eluted.

Achieve high titers of purified adenovirus

The high titer and purity achieved by our Adeno-X purification kits is comparable to that achieved by CsCl density gradient centrifugation, with viral particle-to-infectious unit ratios as low as 20:1. Unlike techniques involving CsCl, our method is safe, and requires no advanced training or expensive equipment—only simple manipulations are required to produce consistently high yields of purified adenovirus.

A powerful pump makes scaling up rapid and easy

To collect large amounts of virus use the Mega kit. With this kit we have adopted the use of a syringe pump (e.g., Genie Plus from Kent Scientific) to drive the clarified lysate through the three-tiered purification cartridge. The consistent pressure and flow rate provided by the pump during the binding, wash, and elution steps provides excellent virus recovery.

Adenovirus is purified directly from the cell pellet of HEK 293 cells. Starting from the second day after infection, the vast majority of virus is found inside the cell, not in the supernatant. Therefore, combining the supernatant with the cell pellet (as with competitor protocols) unnecessarily complicates the protocol without increasing the amount of virus purified.

Adenovirus purification with high yield. HEK-293 cells were seeded and infected with adenovirus and harvested when the cytopathic effect was nearly complete. The resulting adenovirus was then purified according to the Adeno-X Maxi Purification Kit protocol and total IFU was determined for each fraction (crude, flowthrough, wash, and eluate). 72% of the starting material was recovered in the eluate as purified virus. The process was efficient and fast, allowing us to purify 4.68 x 1010 adenoviral particles in one hour.

The Adeno-X Mega Purification Kit protocol

The Adeno-X Mega Purification Kit protocol. From a large-scale culture of infected HEK 293 cells (~25, 15-cm plates), cells are collected and lysed in multiple freeze-thaw cycles. Nucleic acids are degraded to reduce viscosity, and the lysate is clarified and passed through the purification filter assembly. After a brief wash, purified virus is eluted and ready to be used, titered, or stored. The entire process takes less than 1.5 hr.
