Pyrithiamine pPTR vectors
The pyrithiamine or pPTR vectors are E. coli-Aspergillus shuttle vectors and contain an ampicillin resistance gene (AmpR) as the selection marker for E. coli and a pyrithiamine-resistance gene (ptrA) as the selection marker for Aspergillus. Pyrithiamine is a pyridine analog and inhibitor of thiamine. Because the pPTR vector multiple cloning site (MCS) is located within the lacZ region, the presence or absence of target DNA insert can easily be verified using blue/white screening via bacterial plates containing IPTG and X-Gal.
- pPTR I DNA (4,782 bp): chromosomal integration vector for Aspergillus sp.
- pPTR II DNA (10,031 bp): autonomously replicating vector for Aspergillus sp.
- These vectors are supplied in 10 mM Tris-HCl (pH 8.0), 1 mM EDTA

- pPTR I DNA: HindIII, PstI, SmaI, and KpnI
- pPTR II DNA: HindIII, SmaI, and KpnI
Do not use reagents containing thiamine during transformation, as traces of thiamine will decrease the bacterial selection efficiency of pyrithiamine media.
