DsRed-Express and DsRed-Express2 fluorescent proteins

Plasmid vectors for expression of DsRed-Express and DsRed-Express2 fluorescent proteins

DsRed-Express and DsRed-Express2 are so called because they are rapidly maturing variants of Discosoma sp. red fluorescent protein (DsRed). They also demonstrate enhanced solubility (DsRed-Express2 with the higher solubility), reduced green emission, and faster maturation rate than the original DsRed or DsRed2.

Additionally, DsRed-Express fluorescent proteins display a reduced tendency to aggregate (Bevis and Glick 2002). DsRed-Express2 fluorescent protein has been used for whole-cell and stem cell applications (Strack et al. 2008). You may also be interested in mCherry or tdTomato fluorescent proteins.

This lentiviral expression vector encodes a DsRed-Express2 fluorescent protein tag. The protein is a rapidly maturing tetrameric fluorescent protein from Discosoma sp. that has minimal cytotoxicity, high solubility, and exhibits strong and stable expression mammalian cells. Inserting a cDNA in the MCS upstream of the DsRed-Express2 coding sequence joins your protein of interest to the N-terminus of the tag, and allows the fusion protein to be tracked and studied in transduced cells. To package the vector into high-titer, replication-incompetent lentivirus, we recommend using the Lenti-X Packaging Single Shots and the Lenti-X 293T Cell Line. The resulting lentivirus can then be used to transduce virtually any mammalian cell type.

The pRetroX-IRES-DsRedExpress vector is a bicistronic retroviral expression vector containing DsRed-Express as a marker for transfection/infection efficiency. A gene of interest is inserted into the MCS located upstream of the encephalomyocarditis virus (EMCV) internal ribosomal entry site (IRES). The IRES sequence allows the protein of interest and DsRed-Express to be independently translated from the same mRNA transcript. DsRed-Express is a human codon-optimized variant of the Discosoma sp. red fluorescent protein (DsRed) that has been engineered for faster maturation.

Affinity purified recombinant Discosoma sp. red fluorescent protein (rDsRed-Express). rDsRed-Express can be used as a standard for protein gels, Western blots, fluorometry, and fluorescence activated cell sorting (FACS). It can also be used as a control for DsRed-Express expression studies and for microinjection into cells and tissues. At its N-terminus, rDsRed-Express contains a 6xHN epitope tag, used for purification of the protein by metal ion affinity chromatagraphy (IMAC) with our TALON Resin. An enterokinase (EK) site, located between the tag and DsRed-Express, makes it possible to remove the 6xHN polypetide if desired. The 6xHN tag does not interfere with the protein's characteristic spectral properties.

24 DsRedExpress, DsRedExpress2, E2-Crimson, and HcRed are covered by one or more of the following U.S. Patent Nos. 7,157,565; 8,664,471; 8,679,749 and non-U.S. international equivalents in some countries.

pEF1α-IRES-DsRed-Express2 is a bicistronic mammalian expression vector that allows the simultaneous, constitutive expression of a protein of interest and the red fluorescent protein DsRed-Express2. Expression of the bicistronic transcript is driven by the human elongation factor 1 alpha (EF1α) promoter.

Robust expression of DsRed-Express2 in mouse bone marrow hematopoietic stem and progenitor cells

Robust expression of DsRed-Express2 in mouse bone marrow hematopoietic stem and progenitor cells

Robust expression of DsRed-Express and DsRed-Express2 in mouse bone marrow hematopoietic stem and progenitor cells. Mononuclear bone marrow cells were transduced with retroviral vectors encoding DsRed-Express (Panel A), DsRed-Express2 (Panel B), or EGFP (Panel C), and fluorescent cells were sorted 87 hr later. Red and green fluorescence signals were detected using the PE and FITC filter sets, respectively. The lines represent gates defined by analyzing untransduced cells.

Expression of AcGFP1 driven by the EF-1 alpha promoter in stem cell lines is higher than expression driven by the CMV promoter

Expression of AcGFP1 driven by the EF-1 alpha promoter in stem cell lines is higher than expression driven by the CMV promoter

Expression of AcGFP1 driven by the EF-1 alpha promoter in stem cell lines is higher than expression driven by the CMV promoter. The mouse embryonic stem cell lines E14 (Panel A) and D3 (Panel B) were transduced by Lenti-X lentivirus, expressing AcGFP1 either under the control of the CMV promoter or the Elongation factor alpha (EF-1 alpha) promoter. The expression level of AcGFP1 in infected cells five days postinfection was monitored by FACS analysis using the FL1 channel. The expression of AcGFP1 driven by the EF-1 alpha promoter in both stem cell lines was considerably higher compared to the CMV promoter. This is mainly due to a considerably lower rate of silencing of the EF-1 alpha promoter in stem cells compared to the CMV promoter as published (Wang, et al. (2008) Stem Cells Dev 17:279–289).

pEF1α-DsRed-Express2 is a mammalian expression vector that constitutively expresses the red fluorescent protein DsRed-Express2, even after stable integration of the vector into the host cell genome. Stable, constitutive expression of DsRed-Express2 is driven by the human elongation factor 1 alpha (EF1α) promoter, which allows the protein to be expressed without the transgene silencing associated with CMV promoters. The vector, which lacks an MCS, is designed to be used for cell labeling or as a marker of transfection efficiency.
