Takara IVTpro mRNA Synthesis Systems

The Takara IVTpro mRNA Synthesis Systems (Cat. # 6141, 6148 & 6131) offer a complete solution for synthesizing high-quality single-stranded mRNA from your gene of interest, with a cap structure (using CleanCap Reagent AG from TriLink Biotechnology or similar, not included) and poly(A) sequence. They contain:

  • Cloning Kit for mRNA Template (Cat. # 6143 or 6133)—enables easy cloning of a template DNA into a pre-designed In-Fusion cloning-ready plasmid system for in vitro transcription with the Takara IVTpro T7 mRNA Synthesis Kit. The plasmid supplied with Cat. # 6143 contains a Hind III site directly after the poly(A) sequence, while Cat. # 6133 contains a BspQ I site directly after the poly(A) sequence.
  • Takara IVTpro T7 mRNA Synthesis Kit (Cat. # 6144 or 6134)—includes a highly efficient T7 RNA polymerase mix optimized for preparing high yields of mRNA through in vitro transcription. The kit also allows integration with capping systems (not included) and RNA modification with modified NTPs. The Enzyme Mix provided as part of Cat. # 6134 contains PrimeCap T7 RNA Polymerase (low dsRNA) for reduced dsRNA production.

Overview

Takara IVTpro mRNA Synthesis Systems accelerate RNA-focused research with the following benefits:

  • Streamlined workflow—complete in vitro transcription workflow, from template preparation to mRNA purification, with a single kit
  • Ultra-high yield—generate up to 200 µg of mRNA per reaction with a cap structure and poly(A) sequence
  • Flexibility—incorporate a wide variety of capping systems, including CleanCap or ARCA and/or a modified nucleoside such as pseudouridine

More Information

Applications

  • Construction of plasmid DNA for in vitro transcription template
  • Capped mRNA synthesis using TriLink's CleanCap Reagent AG, or other cap analogs such as Anti-Reverse Cap Analog (ARCA)*
  • Synthesis of long non-coding RNA or siRNA precursors for non-coding RNA research
  • Synthesis of RNA standard template for RT-qPCR
  • RNA probe synthesis for hybridization

A different template sequence design is required when using ARCA.
