Isolation of RNA from soil—NucleoBond RNA Soil and NucleoBond RNA Soil Mini

Efficient isolation of up to 600 µg of high-quality RNA from any kind of  soil or sediment sample.

The NucleoBond RNA Soil and NucleoBond RNA Soil Mini kits are designed for the purification of RNA from soil and sediment samples. In combination with the DNA Set for NucleoBond RNA Soil (or DNA Set for NucleoBond RNA Soil Mini), RNA and DNA can be isolated from the same soil sample. Purified nucleic acids are suitable for metagenomic studies, for example, when the genome and transcriptome are analyzed from the same sample. NucleoBond RNA Soil Mini is a downscaled version of the NucleoBond RNA Soil kit.

The NucleoBond RNA Soil and NucleoBond RNA Soil Mini kits are designed for the purification of RNA from soil and sediment samples. In combination with the DNA Set for NucleoBond RNA Soil (or DNA Set for NucleoBond RNA Soil Mini), RNA and DNA can be isolated from the same soil sample. Most soils contain relatively low amounts of RNA, compared to DNA. Therefore, these kits were designed with proven NucleoBond technology to enable variable sample inputs and yield pure nucleic acids. Sample lysis using our NucleoSpin Bead Tubes Type A in the presence of the optimized Lysis Buffer E1 and phenol:chloroform:isoamyl alcohol ensures efficient sample disruption. The optional Buffer OPT enables efficient RNA and DNA purification from mineral and organic soil samples.

For comparison, the samples were processed in parallel using competitor kit "Q" and the corresponding kit protocol. Each RNA sample was eluted in a volume of 100 µl and yields were determined photometrically (left panel). In addition, 15 µl of each eluate was analyzed by gel electrophoresis (1% TAE; right panel). For each sample type, NucleoBond RNA Soil provided a higher yield compared to the competitor kit.

RT-PCR was then performed using 40 µl of eluate from each sample (amplicon size = 466 bp). Samples prepared using NucleoBond RNA Soil demonstrated lower Cp values relative to corresponding samples prepared using kit "Q".

High RNA yields with NucleoBond RNA Soil. Various soil samples (clay, shore soil, and forest soil) were purified in duplicate with NucleoBond RNA Soil using the standard kit protocol. For comparison, the samples were processed in parallel using competitor kit "Q" and the corresponding kit protocol. Each RNA sample was eluted in a volume of 100 µl and yields were determined photometrically (left panel). In addition, 15 µl of each eluate was analyzed by gel electrophoresis (1% TAE; right panel). For each sample type, NucleoBond RNA Soil provided a higher yield compared to the competitor kit.

NucleoBond RNA Soil enables efficient parallel DNA isolation. Nucleic acids purified with these kits are suitable for metagenomic studies. including genomic and transcriptomic analyses. Most soils contain relatively low amounts of RNA compared to DNA. Therefore, this kit was designed to accommodate sample inputs of 0.25–0.5 g in order to yield 0.25–2.5 µg of RNA. Sample lysis using NucleoSpin Bead Tubes Type A in the presence of the optimized Lysis Buffer E1 and phenol:chloroform:isoamyl alcohol ensures efficient sample disruption.
