NucleoBond PC 10000 EF overview

Lysates are cleared by simple filtration using the included NucleoBond Folded Filters—no centrifugation is required.

Endotoxins

Endotoxins (lipopolysaccharides [LPS]) are heat-stable toxins found in the outer membranes of gram-negative bacteria. LPS trigger a strong reaction in mammalian immune systems that can contribute to low-level inflammatory-related diseases or life-threatening states such as septic shock.

Endotoxins can lead to the release of cytokines, such as:   Induced changes due to endotoxins:

  • Tumor necrosis factor
  Cell type Endotoxin
unit (EU)
Effect
  • TNF, IL-1, and IL-10
  Urethral epithelium

25,000

Alters clonal efficiency
  • Anaphylatoxins C5a and C3a
Human T cell 500 Induces proliferation and lymphokine production
  • Myocardial depressant factor
Murine B lymphoma 50 Increases production of light-chain immunoglobulin 30–40 fold
  • Platelet-activating factor
Recombinant CHO 50 Alters production of protein
  • Beta-endorphins
Human IVF embryo 5 Reduces success rates of in vitro fertilization programs 3–4 fold
  • Bradykinin
Aortic ring 5 Reduces cardiac contractility, induces production of IL-1 and tumor necrosis factor
  • Histamine
Equine macrophage 2.5 Induces production of IL-6

Applications

Vaccine production workflow, from viral sequencing to in vitro-transcribed mRNA. NucleoBond PC 10000 EF is used during the culture, amplification, and purification stages to ensure the resulting plasmid DNA is free of harmful endotoxins.

NucleoBond PC 10000 EF can be used in the following applications:

  • Adeno-associated virus (AAV) production
  • Gene therapy research
  • Vaccine development
