Resolving tumor heterogeneity with a fully automated, high-throughput single-cell WGA workflow
With this poster, Dr. Bryan Bell showed how the Shasta™ Whole-Genome Amplification (WGA) automated workflow was used to reliably detect segmental aneuploidies, CNVs, and SNVs in a library of 1,288 single cells. With the Shasta WGA workflow, segmental aneuploidies along with well-characterized chromosomal abnormalities were reliably detected in cell lines and primary tumor samples—such as clear cell renal cell carcinoma (ccRCC)—at a shallow sequencing depth of 2.5 x 105 paired-end reads per cell. The pseudobulk SNV analysis capability aids in identifying tumor subclones even with shallow sequencing depth.
A novel, high-throughput, full gene-body scRNA-seq workflow for improved biomarker discovery
In this poster, Dr. Yue Yun described using the high-throughput Shasta Total RNA-Seq Kit - 2 Chip to construct full gene-body libraries from up to 100,000 cells. In a case study using A549 cells, this technology detected ~11,000 genes and 40,000 transcripts per cell with 100,000 reads per cell. UMAP analysis using either protein-coding or noncoding genes revealed distinct clusters corresponding to different populations or sample types. This technology enhances the capability to identify new biomarkers by enabling comprehensive profiling of both protein-coding and noncoding genes with full gene-body coverage.
