NucleoProtect RNA is a nontoxic, aqueous solution designed to stabilize RNA in mammalian cell or tissue samples for convenient storage prior to RNA isolation (Figure 1). The solution permeates cells and tissues up to 5 mm in diameter. NucleoProtect RNA has been shown to prevent degradation of samples stored at 4°C for up to one month or at or below −20°C for longer periods, eliminating the need for immediate processing or rapid freezing of samples in liquid nitrogen. Best results are obtained if samples are treated with NucleoProtect RNA as soon as possible after harvesting.
RNA stabilization data is shown for samples stored at 4°C for up to one month and room temperature for one week (Figure 2), −75°C for three months (Figure 3), and 37°C for one day (Figure 4). NucleoProtect RNA also provides protection from freeze-thaw damage (Figure 5). RNA from samples stored in NucleoProtect RNA can be isolated using most downstream RNA purification methods, including NucleoSpin RNA kits.
NucleoProtect RNA stabilization solution offers the following benefits:
Compatible with most downstream RNA purification methods, including NucleoSpin RNA kits
Allows for recovery of high-quality RNA with RIN values of 9 or 10
Enables preservation of samples at 4°C for up to one month or at or below −20°C for longer-term storage
Suitable for transport and shipment of cell and tissue samples (no chemical hazard)
Human/animal cells and tissues (max. 5-mm diameter)
Storage time
18–25°C: up to seven days
4°C: up to one month
−20°C/−80°C: long term
Typical RIN after RNA isolation*
9–10 for RNA purified from mammalian tissues and cultured cells, respectively
*Data generated using NucleoSpin kits; RNA integrity strongly depends on quality and handling of samples prior to stabilization.
Applications
Preservation of RNA in animal cells or tissues for short- and long-term storage prior to RNA purification and downstream sample processing
RNA purification, cDNA synthesis, qPCR, NGS

Application data
Figure 2. Reliable stabilization of RNA in samples stored at room temperature or 4°C for one week or one month, respectively, prior to RNA isolation. Using the NucleoSpin RNA Plus kit, RNA was isolated from cell culture (Panel A) and mouse tissue (Panel B) samples that were either untreated (non-stabilized) or stabilized using NucleoProtect RNA (MN) or competitor products (S = RNAlater solution; Z = DNA/RNA Shield). For each sample type and storage condition evaluated, NucleoProtect RNA provided equal or better RNA preservation as compared to the competitor products.
Figure 3. NucleoProtect RNA prevents RNA degradation during sample storage at −75°C. Using the NucleoSpin RNA Plus kit, RNA was purified from various tissue samples that were then stored at −75°C for three months and either unstabilized (non-stabiliz.) or stabilized in NucleoProtect RNA (MN) or RNAlater solution (S). As demonstrated, NucleoProtect RNA effectively prevents degradation of RNA that can occur in unstabilized samples stored at −75°C.
Figure 4. NucleoProtect RNA preserves sample integrity during storage at 37°C. Using the NucleoSpin RNA Plus kit, RNA was purified from tissue samples that were then stored for one day at either 4°C or 37°C in NucleoProtect RNA. Storage in NucleoProtect RNA effectively prevented degradation of RNA even at high ambient temperatures.
Figure 5. Protection from freeze-thaw damage. RNA integrity in samples stabilized with NucleoProtect RNA is well-maintained even after 10 freeze-thaw cycles.
