Retrovirus packaging cell lines and systems

The Retro-X Universal Packaging System is our premium retroviral packaging system featuring the GP2-293 cell line. All four commonly used envelopes are supplied on separate vectors including VSV-G, eco, ampho, and 10A1 to allow you to choose the tropism that is most appropriate for your target cells. The envelope vector is cotransfected with your retroviral expression vector, and high titers of amphotropic, ecotropic, dualtropic, or pantropic virions can be obtained in less than 48 hours.

The Retro-X Universal Packaging System is our premium retroviral packaging system featuring the GP2-293 cell line. All four commonly used envelopes are supplied on separate vectors including VSV-G, eco, ampho, and 10A1 to allow you to choose the tropism that is most appropriate for your target cells. The envelope vector is cotransfected with your retroviral expression vector, and high titers of amphotropic, ecotropic, dualtropic, or pantropic virions can be obtained in less than 48 hours.

RetroPack PT67 Cell Line

RetroPack PT67 is the recommended packaging cell line for creating a stable producer cell line for continuous production of retrovirus. If you wish to make retrovirus via transient transfection, we recommend the Retro-X Universal Packaging System. This retroviral packaging system includes an NIH/3T3-based packaging cell line that expresses the 10A1 viral envelope. Virus packaged from PT67 cells can be used to infect a broad range of mammalian cells including human mouse and rat cell types. The virus is able to enter cells via two different surface molecules, the amphotropic retrovirus receptor (Ram-1) and the GALV receptor.

Pantropic Retroviral Expression System

The Pantropic Retroviral Expression System also features the GP2-293 packaging cell line that expresses gamma-retroviral gag and pol proteins together with pVSV-G. Upon transient transfection of pVSV-G the cell line expresses an envelope glycoprotein from the vesicular stomatitis virus that is not dependent on a cell surface receptor; instead, it mediates viral entry through lipid binding and plasma membrane fusion. Two transfer expression vectors are included with the pantropic system, giving you the choice of using a retroviral 5' LTR or the Drosophila hsp70 promoter to express your gene of interest.

This kit provides a complete retroviral vector packaging system for use with any retroviral vector. It is designed to produce virus that can infect dividing cells from a broad range of mammalian and nonmammalian cell types. The GP2-293 Packaging Cell Line contains only the MoMuLV gag and pol genes. The viral envelope portion of the packaging function (env gene) is supplied by transiently co-transfecting one of the Retro-X Universal Packaging Vectors (p10A1, pAmpho, pEco, or pVSV-G) with the retroviral vector. The pQCLIN Retroviral Vector (included in the vector set) can be used as a positive control to constitutively express β-galactosidase in transduced mammalian cells.

High titer retroviral packaging with a choice of four different retroviral packaging envelopes

High titer retroviral packaging with a choice of four different retroviral packaging envelopes. GP2-293 Cells were transfected with a Retro-X Q vector expressing the red fluorescent protein DsRed2, and each of the four envelope vectors supplied with the Retro-X Unversal Packaging System. After incubating the transfected GP2-293 cells for 48 hr, 10 ?l of the resulting virus was used to infect NIH 3T3 cells. After 48 hours, titers were measured by flow cytometry. Titers were calculated by multiplying the percent infected cells (infectious units) by the total cell number, then dividing by the volume of virus (ml). Panel A. Ecotropic envelope. Panel B.   • We recommend the Retro-X Universal Packaging System for most applications
    • This system produces the highest titers
    • GP2-293 cells are easy to transfect which leads to very high titers
    • It is supplied with all four commonly used envelopes for great flexibility (including VSV-G and Eco)
    • Retrovirus is produced by transient transfection of the envelope vector and retrovirus vector
  • Use RetroPack cells if you wish to create a stable producer cell line
  • All retroviral packaging cell lines and systems are compatible with all retroviral vectors sold by Takara Bio, including all MMLV- and MSCV-based retroviruses.
