High-throughput isolation of genomic DNA from tissue—NucleoSpin 8/96 Tissue

8-well and 96-well preparation of highly pure genomic DNA from animal and human tissue such as mouse and rat tails

NucleoSpin 8/96 Tissue is designed for the rapid and simultaneous preparation of highly pure genomic DNA from animal and human tissue (e.g., mouse and rat tails) for varying throughputs in a flexible 8-well strip format and for high-throughput applications in a 96-well format. Up to 30–40 µg of pure genomic DNA (typical yields are 5–15 µg) can be prepared from a 1.2-cm mouse tail tip section (age of mice is 4–6 weeks). Output DNA can be used directly for PCR, Southern blotting, or any kind of enzymatic reaction.

NucleoSpin 8/96 Tissue is designed for the rapid and simultaneous preparation of highly pure genomic DNA from animal and human tissue (e.g., mouse and rat tails) for varying throughputs in a flexible 8-well strip format and for high-throughput applications in a 96-well format. Up to 30–40 µg of pure genomic DNA (typical yields are 5–15 µg) can be prepared from a 1.2-cm mouse tail tip section (age of mice is 4–6 weeks). Output DNA can be used directly for PCR, Southern blotting, or any kind of enzymatic reaction.

NucleoSpin Tissue Binding Strips/Plates can be used under vacuum or by centrifugation. With the NucleoSpin 8/96 Tissue method, lysis is achieved by incubation of the samples in a solution containing detergents and Proteinase K at 56°C. Appropriate conditions for binding of DNA to the silica membranes of the NucleoSpin Tissue Binding Strip/Plates are created by addition of large amounts of chaotropic ions and ethanol to the lysate. The binding process is reversible and specific to nucleic acids. Contaminants are removed by washing with two different wash buffers. Pure genomic DNA is finally eluted under low ionic strength in a slightly alkaline elution buffer.

The NucleoSpin 96 Tissue Core kit provides the buffers, Proteinase K, and NucleoSpin Tissue Binding Plates only. Accessory plates (e.g., lysis plates, elution plates) are not provided with the core kit. The user can select additional consumables from a variety of suitable accessory plates according to individual needs, for the highest flexibility.
