Isolation of genomic DNA from whole blood—NucleoMag Blood

Genomic DNA extraction from whole blood in 96- or 24-well formats using magnetic bead technology

NucleoMag Blood kits are designed for rapid, parallel purification of genomic DNA from whole blood in 96- or 24-well formats. After lysis of blood cells, genomic DNA is selectively bound to the NucleoMag B-Beads. Contaminants are washed away and the DNA can be eluted from the beads under low-salt conditions. Since the magnetic bead technology provides a closed system (no transfer of sample from one well to another is necessary during the preparation), the risk of cross-contamination is reduced.

NucleoMag Blood kits are designed for rapid, parallel purification of genomic DNA from whole blood in 96- or 24-well formats. After lysis of blood cells, genomic DNA is selectively bound to the NucleoMag B-Beads. Contaminants are washed away and the DNA can be eluted from the beads under low-salt conditions. Since the magnetic bead technology provides a closed system (no transfer of sample from one well to another is necessary during the preparation), the risk of cross-contamination is reduced. NucleoMag technology can be easily adapted to common liquid-handling instruments: it can be used either with static-pin separators (e.g., NucleoMag SEP) or magnetic separators integrated into robotic workstations. Purified DNA obtained with these kits is ready for use in PCR, Southern blotting, or any kind of enzymatic reaction.

High-integrity DNA from whole blood using NucleoMag Blood 200 µL. DNA from human blood (24 individual donors) was extracted. Each sample was extracted in duplicate. The figure shows DNA on a 0.7 % (w/v) TAE agarose gel (ethidium bromide stain). The size standard is λHindIII. The gDNA migrates as a distinct band with a molecular weight >23.5 kb representing the high-integrity of DNA.

NucleoMag Blood 3 mL is designed for rapid, parallel purification of genomic DNA from 3-ml inputs of whole blood in a 24-well format. The kit is compatible with fresh or frozen blood, as well as blood that has been treated with EDTA or citrate, and typically yields up to 100–130 µg of pure genomic DNA per sample. Purified DNA obtained with the kit typically has a A260/A280 ratio ≥1.6–1.9, and is ready for use in PCR, Southern blotting, or any kind of enzymatic reaction.

The NucleoMag Blood 3 mL procedure is based on reversible adsorption of nucleic acids to paramagnetic beads under appropriate buffer conditions. After lysis of blood cells, genomic DNA is selectively bound to the NucleoMag B-Beads. Contaminants are washed away and the DNA can be eluted from the beads under low-salt conditions. Since the magnetic bead technology provides a closed system (no transfer of sample from one well to another is necessary during the preparation), the risk of cross-contamination is reduced. NucleoMag technology can be easily adapted to common liquid-handling instruments: it can be used either with static-pin separators (e.g., NucleoMag SEP) or magnetic separators integrated into robotic workstations. NucleoMag Blood 3 ml is recommended for use with the KingFisher Flex 24 instrument, which enables automated processing of 24 samples in approximately 1 hour.
