Midipreps for plasmid DNA purification—NucleoBond Xtra Midi/Midi Plus

Purify up to 500 µg of ultra-pure plasmid DNA using ion exchange midi prep purification technology in 30 minutes

NucleoBond Xtra Midiprep kits are a new generation of anion-exchange midiprep plasmid purification kits based on patented NucleoBond technology. Typical yields of ≥250 μg of ultra-pure plasmid DNA can be obtained in about half the time compared to other Midiprep kits. The entire plasmid purification procedure takes 68 minutes with NucleoBond Xtra Midi kits and 28 minutes with NucleoBond Xtra Midi Plus kits. Read the NucleoBond Xtra technote to see how it works.

NucleoBond Xtra Midiprep kits are a new generation of anion-exchange midiprep plasmid purification kits based on patented NucleoBond technology. NucleoBond Xtra Midiprep kits contain an enlarged column with a lower silica resin bed. This in turn enables faster flow of lysate and buffers through the columns. Specially designed column filters are included for convenient and time-saving clarification of bacterial lysates. The column filters are supplied inserted in the NucleoBond Xtra midiprep columns and allow parallel clarification of bacterial lysate and loading onto the column. Their large, structured surface leads to high filter-flow rates and minimizes the risk of clogging. The improved silica material has greater DNA binding capacity and is based on the proven NucleoBond anion-exchanger group MAE (methylaminoethanol). Optimized buffer compositions additionally lead to improved alkaline lysis and increased column flow rates.

Reliable plasmid purification from large quantities of cells with NucleoBond Xtra Midi and NucleoBond Xtra Midi EF. Multiple plasmid preps were performed in parallel from bacterial cultures containing varying quantities of cells (ODV200 vs. ODV600) using NucleoBond Xtra Midi, NucleoBond Xtra Midi EF, and the ZymoPURE II Plasmid Midiprep Kit, and analyzed by agarose gel electrophoresis. At both input levels, the NucleoBond Xtra kits provided higher and more consistent yields than the ZymoPURE II kit, and this performance advantage was particularly striking for the higher input amount (ODV400). ODV was calculated by multiplying the optical density of each culture (measured at 600 nm) times the culture volume.

The bacterial lysate is cleared and loaded onto the equilibrated column in one step (plasmid DNA binds to the anion exchange resin)
The bacterial lysate is cleared and loaded onto the equilibrated column in one step (plasmid DNA binds to the anion exchange resin). The first washing step using equilibration buffer is performed with the column filter inserted into the column to wash out residual lysate from the filter and obtain maximum recovery of DNA. After subsequent washing, the purified plasmid DNA is eluted in a high-salt buffer and precipitated with isopropanol. Precipitation can be performed by centrifugation [NucleoBond Xtra Midi (midiprep) or Maxi (maxiprep) kits] or by using the NucleoBond Finalizer.

Higher plasmid yields in less time—in comparison to competitors’ anion exchange-based kits. Plasmid DNA was isolated following each manufacturer's protocol using the maximum culture volume with high plasmid content. The yield of plasmid DNA was determined after DNA precipitation. NucleoBond Xtra Midi/Maxi (midiprep and maxiprep kits) provide up to 60% time savings and up to 100% higher yields compared to competitors’ kits.

Higher yields versus Qiagen with NucleoBond Xtra Midi and NucleoBond Xtra Maxi

Higher yields than Qiagen with NucleoBond Xtra Midi and NucleoBond Xtra Maxi. Plasmid DNA was purified from bacterial cultures using NucleoBond Xtra Midi, NucleoBond Xtra Maxi, and corresponding midiprep and maxiprep kits from Qiagen following the protocols for each kit (in each case, the maximum input volume specified by the corresponding kit protocol was processed). Yields of plasmid DNA were 60–100% higher with the NucleoBond kits than the Qiagen kits (Q Midi = Qiagen Plasmid Midi Kit, Q Maxi = Qiagen Plasmid Maxi Kit).

Reliable plasmid purification from large quantities of cells with NucleoBond Xtra Midi and NucleoBond Xtra Midi EF

Reliable plasmid purification from large quantities of cells with NucleoBond Xtra Midi and NucleoBond Xtra Midi EF. Multiple plasmid preps were performed in parallel from bacterial cultures containing varying quantities of cells (ODV200 vs. ODV600) using NucleoBond Xtra Midi, NucleoBond Xtra Midi EF, and the ZymoPURE II Plasmid Midiprep Kit, and analyzed by agarose gel electrophoresis. At both input levels, the NucleoBond Xtra kits provided higher and more consistent yields than the ZymoPURE II kit, and this performance advantage was particularly striking for the higher input amount (ODV400). ODV was calculated by multiplying the optical density of each culture (measured at 600 nm) times the culture volume.
