Endotoxin-free plasmid midiprep DNA purification—NucleoBond Xtra Midi EF/Midi Plus EF

Transfection grade plasmid purification

NucleoBond PC EF Kits use anion exchange and gravity flow for purification of endotoxin-free plasmid DNA. Endotoxin removal is achieved in a unique washing step—no extra incubations are necessary. Lysates are cleared by simple filtration using the included special column filter, designed for high filter flow rates, and rapid lysate clearing, no centrifugation is required.

NucleoBond PC EF Kits use anion exchange and gravity flow for purification of endotoxin-free plasmid DNA. Endotoxin removal is achieved in a unique washing step—no extra incubations are necessary. Lysates are cleared by simple filtration using the included special column filter, designed for high filter flow rates, and rapid lysate clearing, no centrifugation is required. Additionally NucleoBond Xtra has improved silica material with higher binding capacity, leading to typical yields of 400 μg (Midi) and 1000 μg (Maxi). 

NucleoBond Xtra Finalizers: The finalizers, provided with the Plus kits, replace the time-consuming centrifugation step that follows every isopropanol precipitation of diluted DNA samples.

Reliable plasmid purification from large quantities of cells with NucleoBond Xtra Midi and NucleoBond Xtra Midi EF. Multiple plasmid preps were performed in parallel from bacterial cultures containing varying quantities of cells (ODV200 vs. ODV600) using NucleoBond Xtra Midi, NucleoBond Xtra Midi EF, and the ZymoPURE II Plasmid Midiprep Kit, and analyzed by agarose gel electrophoresis. At both input levels, the NucleoBond Xtra kits provided higher and more consistent yields than the ZymoPURE II kit, and this performance advantage was particularly striking for the higher input amount (ODV400). ODV was calculated by multiplying the optical density of each culture (measured at 600 nm) times the culture volume.
