Multiple tissue cDNA panels

MTC multiple tissue cDNA panels are sets of first-strand cDNAs from 5 to 12 different tissues or cell lines. They offer fast and accurate PCR analysis of gene expression across multiple tissues in a fraction of the time required for Northern or dot blot analysis. Each MTC panel has been normalized to several different housekeeping genes to ensure accurate assessment of target mRNA abundance. In addition, human panels are normalized against one another, allowing comparison of results obtained using different panels. MTC panels are virtually free of genomic DNA, ensuring all signals generated are from actual mRNA transcripts.

MTC multiple tissue cDNA panels are sets of first-strand cDNAs from 5 to 12 different tissues or cell lines. They offer fast and accurate PCR analysis of gene expression across multiple tissues in a fraction of the time required for Northern or dot blot analysis. Each MTC panel has been normalized to several different housekeeping genes to ensure accurate assessment of target mRNA abundance. In addition, human panels are normalized against one another, allowing comparison of results obtained using different panels. In addition to two broad-coverage human cDNA panels, we offer human digestive system, immune system, and cell line-specific panels. For rapid screening of gene expression under different apoptotic conditions we provide an MCF7 Apoptosis cDNA Panel, consisting of first-strand cDNAs synthesized from total RNA of cells treated with various apoptosis-inducing agents. The MCF7 cDNA Panel, designed for studies focusing on p53-regulated apoptosis, contains cDNA from modified and unmodified MCF7 cells expressing dominant-negative p53.

Human cardiac beta-myosin heavy chain fragments of different lengths were amplified from 100 ng of genomic DNA using Titanium Taq and two leading competitor’s hot start Taq polymerases. Optimal conditions were used for each enzyme, as specified by the manufacturer. Lane 1: 0.5 kb fragment. Lane 2: 1 kb fragment. Lane 3: 1.8 kb fragment. Lane M: 1 kb DNA size ladder.

TITANIUM Taq is active over a wide range of Mg2+ concentrations. TITANIUM was used to amplify a 500 bp region of Calf Thymus genomic DNA. The MgCl2 concentration was varied as indicated. The enzyme performed consistently through the range of Mg2+ used. Lane M: DNA size marker.

G3PDH PCR Primers and Control cDNA are included for 10 positive control reactions.

Tissue expression patterns of two mRNAs in MTC Human Panels I & II. Panel A. A 328-bp fragment was amplified in 35 cycles from Human Panel I using Delta lactoferrin (Lf) primers. Panel B. A 983-bp fragment was amplified in 21 cycles from Human Panel II using GAPDH primers. The PCR products were then electrophoresed on 2% agarose/EtBr gels. Data is shown for 16 samples from each of the two panels. Lane 17: negative control PCR (no cDNA).
