Capturem high-capacity membranes

Capturem nickel-functionalized membranes for his-tagged purification are a revolutionary solution for recombinant protein purification that effortlessly outperforms conventional methods. Novel nylon membranes have been assembled into spin columns and filtration devices, creating no-waiting, room-temperature workflows (just 5–30 minutes from loading a sample through elution) compatible with both mammalian and bacterial cells.

Capturem nickel-functionalized membranes for his-tagged purification are a revolutionary solution for recombinant protein purification that effortlessly outperforms conventional methods. Novel nylon membranes have been assembled into spin columns and filtration devices, creating no-waiting, room-temperature workflows (just 5–30 minutes from loading a sample through elution) compatible with both mammalian and bacterial cells. Exceptional performance is consistent under a wide range of research settings, including denaturing conditions, in the presence of additives (e.g., BME, EDTA, DTT, glycerol, TCEP, etc.; see compatibility table), and when performing tandem purification of his-tagged proteins.

The modified nylon membranes have a small bed volume, yet still produce high-purity, concentrated protein, due to the chemically enhanced surface area of the pores. With this technology, protein-binding capacity is comparable to, or better than, resins that bind 75 mg or more per cm3 of membrane. Miniprep, maxiprep, 24-well-plate, 96-well-plate, and large-volume formats are available to accommodate a range of input volumes. Furthermore, the short residence time of protein samples greatly reduces the possibility of protein degradation or loss of activity, giving downstream applications a higher probability of success.

Protein purification miniprep workflow
Protein purification miniprep workflow. Each mini spin column can be loaded with up to 800 μl of lysate (yielded from 2–5 ml of culture). His-tagged protein is first bound to the membrane, followed by washing with 300 μl wash buffer, and elution with 300 μl elution buffer. Over 90% of the bound protein can be eluted with as little as 100 μl elution buffer. Each step is followed by spinning the tube for 1 min at 11,000 x g. The working bed volume of the membrane is <2 μl. This entire purification is complete in <5 min.

Cells were lysed, and the clarified lysate was loaded onto an equilibrated column. The column was washed with 400 μl wash buffer, followed by elution with 300 μl elution buffer. The Capturem His-Tagged Purification Large Volume unit lets you quickly purify up to 25 mg of protein (depending on protein size and expression level) from a large volume of lysate (up to 500 ml at a time), resulting in purified protein concentrated at up to 2 mg/ml.
