The Lenti-X p24 Rapid Titer Kit (Single Wash) and the Lenti-X p24 Rapid Titer Kit are designed for the rapid determination of lentiviral supernatant titers by p24 ELISA. (See the workflow comparison for both the kits.)

The Lenti-X p24 Rapid Titer Kit (Single Wash) and the Lenti-X p24 Rapid Titer Kit are designed for the rapid determination of lentiviral supernatant titers by p24 ELISA.

What is p24?

Recombinant lentiviruses generated for research use contain major structural proteins, including envelope proteins, matrix proteins, and virus core proteins. The gag gene encodes the viral capsid protein (p24), the nucleocapsid proteins (p6 and p7), and a matrix protein (p17) (see data images by expanding the Details tab in the product table). The Lenti-X p24 Rapid Titer Kit specifically measures the amount of p24 capsid protein present in your viral supernatant. The level of p24 correlates directly with virus titer.

The Lenti-X p24 Rapid Titer Kit (Single Wash) allows you to quickly determine the titer of any HIV-1-based lentiviral vector supernatant using a fast and easy ELISA-based protocol. The test samples are mixed with anti-p24 antibody, HRP-conjugated secondary antibody, and lysis buffer. This mix is then added to the wells of the included microtiter plate (12 x 8-well strips) which are coated with anti-HIV-1 p24 capture antibody and binds the p24 (an HIV-1 core protein) in your test samples. After a single wash step, the specifically-bound p24 in the sample is detected with a color-producing substrate. Color intensity is measured spectrophotometrically to indicate the level of p24 in the samples, which is then quantified against a p24 standard curve in order to titrate the viral content.

Workflow. Lentiviral p24 core protein in packaging cell supernatants is bound to wells of a microtiter plate coated with p24 capture antibody. The presence of bound p24 in the wells is detected using an anti-p24 antibody, HRP-conjugated secondary antibody, and a color-producing substrate. Quantitation is performed by comparing test samples to a p24 standard curve.

The Lenti-X p24 Rapid Titer Kit allows you to quickly determine the titer of any HIV-1-based lentiviral vector supernatant using a standard ELISA. The wells of the included microtiter plate (12 x 8-well strips) are coated with anti-HIV-1 p24 capture antibody, which binds the p24 (an HIV-1 core protein) in your test samples. Specifically-bound p24 is detected in a typical "sandwich" ELISA format using a biotinylated anti-p24 secondary antibody, a streptavidin-HRP conjugate, and a color-producing substrate. Color intensity is measured spectrophotometrically to indicate the level of p24 in the samples, which is then quantified against a p24 standard curve in order to titrate the viral content.
