BcaBEST Isothermal FluorDetect Kit (DNA/RNA)

Rapidly detect DNA or RNA targets without needing to switch reagents or kits. The BcaBEST Isothermal FluorDetect Kit (DNA/RNA) enables fluorescent detection of nucleic acid sequences using either loop-mediated isothermal amplification (LAMP) or one-step RT-LAMP.

Rapidly detect DNA or RNA targets without needing to switch reagents or kits. The BcaBEST Isothermal FluorDetect Kit (DNA/RNA) enables fluorescent detection of nucleic acid sequences using either loop-mediated isothermal amplification (LAMP) or one-step RT-LAMP.

The core of the kit is the BcaBEST Isothermal Mix, an optimized premix containing BcaBEST DNA Polymerase ver. 2.0 and PrimeScript III Reverse Transcriptase. This allows seamless amplification from both DNA and RNA templates. Detection is achieved through TB Green Solution, which enables real-time fluorescence monitoring using a PCR system equipped with a FAM filter. BcaBEST Isothermal Mix also includes Uracil-N-Glycosylase (UNG) to prevent false positives from carryover contamination.

To run the assay, simply combine your DNA or RNA template with standard LAMP primers, the BcaBEST Isothermal Mix, and TB Green Solution. Incubate the reaction at 63°C for 20 minutes using a real-time PCR system to detect and quantify amplification.

BcaBEST Isothermal FluorDetect Kit (DNA/RNA) achieved rapid detection of DNA and RNA samples. BcaBEST Isothermal FluorDetect Kit (DNA/RNA) was used to amplify DNA and RNA samples (template amount = 1, 10, 100, or 1,000 pg or 5, 50, 500, or 5,000 copies) at n = 2. The time to reach a constant fluorescence intensity was measured. For these samples, detection was generally achieved within 20 min. The "1/2" in the figure indicates that one of the n = 2 was not detected within 30 min. For the NTC (no-template control), no amplification within 30 min was observed except for one NTC for orf-134. RNA samples: HL-60 total RNA (ACTB and ALAS1) and SARS-CoV-2 RNA controls (Twist Bioscience). DNA samples: E. coli genomic DNA (fecA) and lambda phage genomic DNA (orf-134).

BcaBEST Isothermal FluorDetect Kit (DNA/RNA) and fluorescent LAMP detection reagents from various companies were used to amplify HL-60 total RNA (ACTB; template amount 1, 10, 100, or 1,000 pg) at n = 2. The time to reach a certain fluorescence intensity was measured. The BcaBEST Isothermal FluorDetect Kit (DNA/RNA) was capable of detection within 15 min.

BcaBEST Isothermal FluorDetect Kit (DNA/RNA) and fluorescent LAMP detection reagents from various companies were used to detect SARS CoV-2 (noninfectious; 1,000 copies/rxn) spiked into nasopharyngeal fluid (amount = 0, 0.1, or 1 μl/rxn) at n = 2, Chlamydia trachomatis (noninfectious; 180 copies/rxn) spiked into urine (amount = 0, 2.25, or 4.5 μl/rxn) at n = 3, and Neisseria gonorrhoeae (noninfectious; 180 copies/rxn) spiked into urine (amount = 0, 2.25, or 4.5 μl/rxn) at n = 3. The spiked samples were treated with Proteinase K and then heat inactivated before the assay. The time to reach a certain fluorescence intensity was measured.

BcaBEST Isothermal FluorDetect Kit (DNA/RNA) is compatible with Q-Probe detection. BcaBEST Isothermal FluorDetect Kit (DNA/RNA) was used to amplify Chlamydia trachomatis control DNA (template amount = 1,000 or 200 copies/rxn at n = 2 and 0 copies/rxn at n = 6) and Neisseria gonorrhoeae control DNA (template amount = 1,000 or 200 copies/rxn at n = 2 and 0 copies/rxn at n = 6). The time for the fluorescence of TB Green (intercalator) or Q-Probe to reach the threshold were measured. Q-Probe hybridizes with amplified LAMP products, indicating decreasing fluorescence. The Q-Probe exhibits the same detection time as the intercalator but demonstrates more specificity in this assay.
