The response vector supplied with these Tet-On 3G tetracycline-inducible expression systems express two transgenes from our premium TRE3G inducible promoter (PTRE3G). You can either co-express two different genes of interest using systems that contain the pTRE3G-IRES Vector, which contains two multiple cloning sites flanking an IRES sequence (Cat. # 631166 and 631346), or you can monitor co-induced expression of your transgene with a fluorescent protein using kits that contain pTRE3G-mCherry (Cat. # 631165 and 631347) or pTRE3G-ZsGreen1 (Cat. # 631164 and 631348).
These IRES-containing bicistronic vectors allow the simultaneous induced expression of two proteins separately but from the same RNA transcript.
How does an IRES work?
The IRES of the encephalomyocarditis virus (ECMV) permits the translation of two open reading frames from one messenger RNA. Although translation initiation of eukaryotic mRNAs occurs almost exclusively at the 5' cap, the IRES allows ribosomes to bind and initiate translation at a second, internal location. Thus, two proteins are expressed simultaneously from the same bicistronic mRNA transcript.
