PrimePath Direct Saliva SARS-CoV-2 Detection Kit

PrimePath Direct Saliva SARS-CoV-2 Detection Kit is a research-use-only, complete kit for detecting SARS-CoV-2 directly from saliva specimens without the need for any DNA/RNA extraction. The workflow has been thoroughly formulated to ensure safe sample handling, reduce hands-on time, and deliver faster, more sensitive results.

PrimePath Direct Saliva SARS-CoV-2 Detection Kit is a research-use-only, complete kit for detecting SARS-CoV-2 directly from saliva specimens without the need for any DNA/RNA extraction. The workflow has been thoroughly formulated to ensure safe sample handling, reduce hands-on time, and deliver faster, more sensitive results.

The kit contains all components required to collect the saliva specimens and run the RT-qPCR detection assay, including our high-performance One-Step PrimeScript III RT-qPCR Kit (Cat. # RR600A). This assay is based on primers and probes designed and published by the CDC.

One Step PrimeScript III RT-qPCR Mix is a dedicated reagent for one-step real-time, probe-based RT-qPCR (using the 5’ nuclease method). This 2X premix does not freeze at its storage temperature of –20℃, so a reaction can be started simply by adding the template sample, primer, and a probe for detecting the desired target. The quick and simple protocol allows the reverse transcription and qPCR reactions to be performed in the same tube. The reverse transcription reaction uses the novel PrimeScript III RTase, which displays increased heat tolerance (up to 55℃) while maintaining the specificity and extensibility of PrimeScript RTase. This allows cDNA synthesis from RNA with a more complex secondary structure. After cDNA synthesis, TaKaRa Taq HS performs highly specific and efficient PCR amplification, while the fluorescence emitted by the probe is detected in real-time. One Step PrimeScript III RT-qPCR Mix is also highly resistant to a wide variety of inhibitory substances in blood and soil, allowing stable one-step real-time RT-qPCR to be performed on a wide range of samples. This product can be used for various applications such as gene expression, RNA virus detection, etc.

Cat. # RR600A contains sufficient reagent for performing 200 reactions in a volume of 25 µl per reaction.

The PrimePath Saliva Collection Kit contains collection sticks and a viral inactivation buffer that is used both for storage and processing samples. At one end of each collection stick, there are channels for collecting liquid such as saliva. When mixed with the collected saliva sample, the buffer will inactivate any SARS-CoV-2 particles that are present. This process makes the saliva compatible with the RT-qPCR step in the PrimePath Saliva Direct SARS-CoV-2 Detection Kit.

Detection of the SARS-CoV-2 nucleocapsid gene by qPCR
PrimePath SARS-CoV-2 Assay Mix was compared to that of Competitor E in a quantitative PCR (qPCR) assay. For both the N1 (FAM) and N2 (HEX) regions of the SARS-CoV-2 nucleocapsid gene, our reagent delivered greater fluorescence intensity, enabling easier detection of the pathogen.

Both the N1 (FAM) and N2 (HEX) regions of the SARS-CoV-2 nucleocapsid gene were quantified using droplet digital PCR (ddPCR). At all dilutions tested, the copy number of each target was calculated to be a consistent value.

Overview

  • Complete kit—all components needed for the user to perform SARS-CoV-2 detection are included (except the qPCR instrument). The kit includes our high-performance One Step PrimeScript III RT-PCR Kit for a simple setup. The in vitro-transcribed (IVT) PrimePath SARS-CoV-2 Positive Control is validated using ddPCR and comprises exact copies of the target RNA sought by the CDC-designed primers and probes in this assay.
  • Easier, safer sample collection and handling—PrimePath Saliva Collection Kit provides unique collection sticks for easy capture of saliva samples. Additionally, the kit comes with a buffer that inactivates SARS-CoV-2 once the saliva sample is dissolved, making it safer for handling and transportation.
  • Faster results—no DNA/RNA extraction is necessary. A total qPCR assay detecting SARS-CoV-2 can be completed in <30 minutes.
