PicoPLEX WGA technology

Whole-genome amplification kits

PicoPLEX technology has long been regarded as the gold standard for whole-genome amplification (WGA) by virtue of the unprecedented reproducibility it provides for analyses involving single-cell or ultra-low inputs. We currently offer three different standalone versions of PicoPLEX technology for WGA directly from 1–10 cells or genomic DNA equivalents. This technology can be paired with NGS library preparation for DNA-seq applications or incorporated into array or qPCR-based approaches.

The respective versions of PicoPLEX WGA technology (and associated applications) are described below:

PicoPLEX WGA Kit (Cat. # R30050):

The PicoPLEX WGA Kit represents the original iteration of PicoPLEX technology (PicoPLEX v1) and is employed in a broad range of different assays for detection and measurement of copy number variations (CNVs), including preimplantation genetic testing for aneuploidy (PGT-A). The PicoPLEX WGA Kit provides superior reproducibility and coverage uniformity versus competitor technologies and is specifically recommended for use in established assays that were developed and/or validated using v1 PicoPLEX WGA technology.

PicoPLEX Single Cell WGA Kit (Cat. # R300671, R300672, R300673):

The PicoPLEX Single Cell WGA Kit (PicoPLEX v2) includes refinements that provide improved evenness and reproducibility of amplification and higher yields relative to its predecessor. From this standpoint, the kit is recommended for development/application of new assays for CNV analysis.

PicoPLEX Single Cell WGA Kit v3 (Cat. # R300718, R300722, R300723):

The PicoPLEX Single Cell WGA Kit v3 (PicoPLEX v3) incorporates a high-fidelity polymerase and additional refinements to allow for more accurate calling of single-nucleotide variants (SNVs) and reduced rates of allele dropout. This kit is well-suited for applications requiring base-level resolution, including detection of SNVs, while it also provides excellent performance for CNV analysis. In addition to providing PicoPLEX v3 technology in a standalone WGA kit, it is also incorporated into the PicoPLEX Gold Single Cell DNA-seq Kit, which provides a comprehensive solution for WGA and NGS library prep.

The PicoPLEX Single Cell WGA Kit uses a single-tube protocol developed specifically for reproducible amplification of single-copy genomic DNA (gDNA) starting with 1–10 cells or equivalent picogram quantities of isolated gDNA. Cell lysis and whole-genome preamplification are followed by amplification, which results in libraries with very low background and typical yields of 8–12 µg of product in under 3 hours. This system is best suited for applications in which high yield is most important. This product contains reagents for 24 reactions.
