PicoPLEX Gold DNA-Seq

The PicoPLEX Gold Single Cell DNA-Seq Kit (PicoPLEX Gold) is designed to generate high-quality DNA libraries from single cells for sequencing on Illumina platforms. The kit is based on our patented PicoPLEX technology and is optimized for the reproducible detection of copy number variants (CNVs), single nucleotide variants (SNVs), indels, and small structural variants. The PicoPLEX Gold kit outperforms the leading Multiple Displacement Amplification (MDA) technology for SNV detection.

Please note that the PicoPLEX Gold Single Cell DNA-Seq Kit has been reconfigured and no longer includes indexes. We offer compatible index kits in plate format (for multiplexing up to 384 samples) or tube format (for multiplexing up to 96 samples). Click the "More" link below for a full description of compatible indexing kits.

Our indexing kits are described below.

PicoPLEX WGA (PicoPLEX) quasi-random priming technology is the international gold standard for whole genome amplification (WGA) for subsequent detection of CNVs in fixed or unfixed single cells. Initially, PicoPLEX chemistry was created for the reproducible detection of aneuploidies and CNVs. To address the need for accurate detection of SNVs, we further optimized the PicoPLEX WGA chemistry using enzymes, primers, and protocols that improve sequencing coverage, uniformity, and accuracy while increasing the resolution for CNVs, SNVs, indels, and other small structural variants while retaining reproducibility.

PicoPLEX Gold provides enhanced PicoPLEX WGA chemistry in the context of a simple, four-step protocol to convert fixed or unfixed single cells into NGS libraries in under three hours with minimal hands-on time.

Unique Dual Index kits (plate format)

Unique Dual Index kits are designed for use with the vast majority of NGS library preparation kits from Takara Bio for multiplexed sequencing on Illumina sequencers. These kits contain indexed PCR primers that include the same adapter as the IDT for Illumina-TruSeq DNA and RNA UD Indexes, with Cat. # 634752 and 634756 carrying the “IDT for Illumina” indexes and Cat. # 634753–634755 carrying indexes that have been developed and validated by Takara Bio. These kits offer a total of 384 dual indexes for multiplexing of up to 384 samples. The indexed PCR primers are supplied pre-dispensed in five different 96-well plate formats, four of which contain one-fourth of the possible dual index combinations (Cat. # 634752–634755) with the remaining kit (Cat. # 634756) containing 24 dual index combinations that are a subset of Cat. # 634752. Each dual index combination is provided at a volume of 5 µl and intended for a single use.

DNA Unique Dual Index kits (tube format)

DNA Unique Dual Index kits are designed for use with ThruPLEX and PicoPLEX library preparation kits to construct libraries for multiplexed sequencing on Illumina sequencers. These kits contain indexed PCR primers carrying the "IDT for Illumina UD" index sequences and offer a total of 96 dual indexes for multiplexing of up to 96 samples. The indexed PCR primers are supplied pre-dispensed in four different sets of 24 individual tubes, each of which contains one-fourth of the possible dual index combinations (Cat. # R400665–R400668). Each dual index tube (24U Sets A–D) contains sufficient volume for up to two uses.

DNA Single Index kits (tube format)

DNA Single Index kits are designed for use with ThruPLEX and PicoPLEX library preparation kits to construct libraries for multiplexed sequencing on Illumina sequencers. These kits contain indexed PCR primers carrying the "TruSeq LT set A" index sequences and offer a total of 24 single indexes for multiplexing of up to 24 samples. The indexed PCR primers are supplied pre-dispensed in two different sets of 12 individual tubes (Cat. # R400695 and R400697), each containing a different index sequence. Each single index tube contains sufficient volume for up to eight uses.
