SMART-Seq PLUS kits: reliable and complete workflows from cDNA through library preparation

  • Streamlined workflows from a single provider give users a seamless, easy experience and reproducible data
  • cDNA generation solutions for either cells—including single cells—or bulk RNA (up to 10 ng)
  • Flexible cDNA input means no required dilution of the generated libraries before sequencing
  • High library yields allow for the flexibility to sequence more deeply
  • Unique dual indexes (UDIs) available for increased confidence when sequencing on the NovaSeq™ system

Proven cDNA synthesis: SMART technology

Begin your sample prep with confidence using our patented SMART (Switching Mechanism at 5′ End of RNA Template) technology to generate cDNA. This technology ensures that the final cDNA libraries contain the entire length of the mRNA—including the 5' end—maintaining a true representation of the original mRNA transcripts, which is a critical factor for gene expression analysis. Start from our: SMART-Seq Single Cell Kit to generate cDNA from single cells, SMART-Seq v4 Ultra Low Input RNA Kit for Sequencing to generate cDNA from multiple intact cells (up to 1,000 cells) or 10 pg–10 ng of total RNA, or SMART-Seq HT Kit for automation-friendly generation of cDNA from single cells or 10 pg–1 ng of total RNA.

Simple library prep workflow: one tube and done

Addition-only library preparation steps provide a faster, failproof experience without lengthy bead purifications that may cause sample mix-up or sample loss and impact data quality. Without hidden steps, the process is easy to automate, scale, and standardize. Preplated, single-use UDIs make assignments clear for your precious samples.

SMART-Seq PLUS Library Prep kit workflow

Figure 1. The SMART-Seq PLUS kits' library preparation methods feature fast, simple workflows without bead purification after ligation—saving time, money, and resources while providing increased confidence in the data.

Higher library yields

Compared to the gold-standard library preparation method, our SMART-Seq PLUS kits generate higher library yields when starting from SMART-Seq cDNA. Higher library yield allows for sequencing on high-throughput sequencers (such as the NovaSeq system) with ease, and excess yield can be used to perform subsequent sequencing runs if desired. Combined with UDIs, you can confidently pool multiple libraries and sequence on one sequencer, saving time and resources.

SMART-Seq PLUS kits generate higher library yields than Nextera XT when using the same cDNA input.

Figure 2. Generate higher library yield to allow for sequencing on high-throughput sequencers with ease.

Identify a greater number of genes

Additionally, our SMART-Seq PLUS kits—especially the SS-HT PLUS kit—capture a higher number of genes compared to the gold-standard library preparation method, indicating a greater capture of the original starting material. Be confident knowing that you can learn more about your precious samples.

SMART-Seq PLUS libraries identify more genes than Nextera XT.

Figure 3. Detect a greater number of genes due to greater capture of the starting material from the cDNA input.

The SMART-Seq Single Cell PLUS Kit demonstrates greater discrimination between control and single-cell samples than NEBNext.

The SMART-Seq Single Cell PLUS Kit demonstrates greater discrimination between control and single-cell samples than NEBNext.

Clustering of the SSsc single cell and positive controls and significantly distinct discrimination of the negative controls provides confidence in the data generated with SSsc PLUS. By UMAP analysis, there is a clear separation between the negative controls and the positive controls/single-cell samples for SSsc PLUS. As expected, the positive controls (grey) and the single-cell samples (green) cluster together for the SSsc PLUS while the negative controls (dusty blue) cluster together near the bottom of the chart. Interestingly, the SSsc PLUS negative controls and all of the NEBNext samples cluster together: negative (red), positive (light blue), and the single cells (purple). These analyses indicate that experimental noise, even when working with such small amounts of starting material, will not affect confidence in the biological import of the results. Data is shown for SMART-Seq Single Cell PLUS (SSsc PLUS). SMART-Seq mRNA Single Cell LP is an equivalent replacement for SSsc PLUS.
