GS2-M medium: maintenance of germline-competent mouse and human ES and iPS cells

GS2-M medium

GS2‐M is a completely defined, serum‐free cell culture media formulation for the generation and long-term maintenance of human and mouse embryonic stem (ES) and induced pluripotent stem (iPS) cell lines. The medium contains two selective small molecule inhibitors that block differentiation‐inducing signals and promote cell survival. 

GS2‐M is a completely defined, serum‐free cell culture media formulation for the generation and long-term maintenance of human and mouse embryonic stem (ES) and induced pluripotent stem (iPS) cell lines. The medium contains two selective small molecule inhibitors that block differentiation‐inducing signals and promote cell survival.

GS2-M medium can be used in combination with the self-renewal cytokine leukemia inhibitory factor (LIF) to convert partial or pre‐iPS cells from mouse into fully pluripotent iPS cells via upregulation of Nanog (Silva et al. 2008; Silva et al. 2009).

GS2-M is a proprietary, defined and serum-free cell culture media formulation for the generation of mouse ES and iPS cell lines in the presence of two selective small molecule inhibitors CHIR99021 and PD0325901 ('2i') that act to eliminate differentiation-inducing signals from GSK3β and ERK/MEK, respectively, and promote cell survival. LIF can also be added to orchestrate the efficient conversion of partial or pre-iPS mouse cells into fully pluripotent iPS cells via Nanog. Long term maintenance of these naïve, 'ground state' mouse pluripotent stem cells is then solely dependent on GS2-M alone.

The ground-state pluripotency marker Rex1 was analyzed via flow cytometry in a Rex1-GFP transduced mouse embryonic stem cell line. Prior to analysis, cells were cultured in either GS2-M or 10%FBS/GMEM + LIF for 3 passages. Cells grown in GS2-M exhibited superior ground-state maintenance.
