The Lenti-X Expression System enables you to produce exceptionally high titers of recombinant lentivirus for expression of any cDNA in any cell type susceptible to lentivirus transduction. The Lenti-X packaging system is a proprietary combination of plasmids that collectively expresses all the viral proteins necessary for high-efficiency packaging of lentiviral vector transcripts into infectious lentivirus particles. Lenti-X Packaging Single Shots, which contain lyophilized Lenti-X packaging plasmids and transfection reagent, are used for efficient introduction of the packaging mix and the pLVX-Puro Vector into 293T cells with very high efficiency. The concerted effects of these components allow 293T cells to produce high-titer replication incompetent lentiviral supernatants for a safe transduction of virtually any cell type. The Lenti-X GoStix can be used to determine whether virus production is within a usable range or for selecting the best time to harvest your virus. The system provides enough reagents for 16 packaging reactions.

The Lenti-X 293T Cell Line is a subclone of the transformed human embryonic kidney cell line, HEK 293, which is highly transfectable and supports high levels of viral protein expression. When transfected with Lenti-X Packaging Single Shots and a lentiviral vector, these cells are capable of producing lentiviral titers as high as >108 ifu/ml, as determined by flow cytometry. The cell line also constitutively expresses the simian virus 40 (SV40) large T antigen.

This kit provides a complete retroviral vector packaging system for use with any retroviral vector. It is designed to produce virus that can infect dividing cells from a broad range of mammalian and nonmammalian cell types. The GP2-293 Packaging Cell Line contains only the MoMuLV gag and pol genes. The viral envelope portion of the packaging function (env gene) is supplied by transiently co-transfecting one of the Retro-X Universal Packaging Vectors (p10A1, pAmpho, pEco, or pVSV-G) with the retroviral vector. The pQCLIN Retroviral Vector (included in the vector set) can be used as a positive control to constitutively express β-galactosidase in transduced mammalian cells.

The System contains an HIV-1-based, lentiviral expression vector, pLVX-EF1α-IRES-Puro, designed to constitutively express a protein of interest and puromycin resistance (Puror). Constitutive expression of these proteins is driven by the human elongation factor 1 alpha (EF1α) promoter, which allows protein expression in mammalian cells in which the CMV promoter is silenced. The System also includes Lenti-X Packaging Single Shots (VSV-G) and Lenti-X GoStix.
