T-Vector pMD20 is a linearized vector with a single 3'-terminal thymidine at both ends. The T-overhang ends at the cloning site improves the efficiency of ligation of PCR products which contain A-overhangs at 3'-ends. T-Vector pMD20 is derived from a pUC19 backbone with modifications in the coding sequence to reduce the false positive rate and improve the positive colonies screened by blue/white color on ampicillin/IPTG/X-Gal LB plates. In addition, an SP6 promoter has been inserted upstream of the multiple cloning site. Therefore, the inserted DNA fragments can be transcribed by SP6 RNA Polymerase (Cat. #2520). T-Vector pMD20 is supplied with a Control Insert (500 bp) DNA for positive control reactions.

T-Vector pMD19 (Simple) is a linearized vector with a single 3'-terminal thymidine at both ends. The T-overhang ends at the cloning site improve the efficiency of ligation of PCR products which contain A-overhangs at 3' -ends. The multiple cloning sites in the lac Z gene sequence have been deleted from this vector, but its β-galactosidase activity is not disrupted. Therefore, transformant colonies containing recombinant plasmids can be identified by blue/white screening. Due to lack of multiple cloning sites in the vector, the inserted PCR product cannot be recut using restriction enzymes. When the insert DNA fragment is recut from the vector, the PCR primers should be used to add a restriction site to the 5'-end for restriction enzyme digestion and subcloning. T-Vector pMD19 (Simple) is supplied with a Control Insert (500 bp) DNA for positive control reactions.

Takara's DNA Ligation Kit, Mighty Mix, is a one-solution premix ligation reagent that offers high efficiency ligations, particularly for blunt-ended ligation. The reaction can be performed for 5 minutes at 25°C (rapid protocol), or 30 minutes at 16°C (standard protocol) with equal results for both circular and linear DNA ligations. The 2X Ligation Mix allows small ligation reaction volumes (10 μl) when DNA is limiting. The kit includes sufficient reagent for 100 ligation reactions. Ligation products reaction can be used directly in bacterial transformation.
