Even if your lab does reprogramming, creating hiPS cell lines can be an inefficient process with heavy costs of time and resources. Additionally, creating an iPS cell line with your specific requirements is difficult due to the challenges inherent to the sourcing process. To make your iPS cell research easier on your team, we offer a reprogramming package to create iPS cells from your (or sourced) human PBMCs. If you already have your own fibroblasts, we can also reprogram them for you.

With Cellartis reprogramming services, we will deliver high-quality human iPS cell lines from your PBMC or fibroblast samples. The cells will be reprogrammed using footprint-free Sendai virus technology to ensure no viral integration.
The reprogramming service can start with sending us your cell samples, or alternatively, you may elect to use Cellartis sourcing services to help you find exactly the right PBMCs to create a custom iPS cell line. Once the reprogramming service begins, the starting material will be prepared for reprogramming; one iPS cell clone will be established in the Cellartis DEF-CS 500 Culture System, then expanded into a stable iPS cell line, banked, and characterized.

Cellartis Descriptive Karyotyping Service (Cat. #Y60304)
Karyotype analysis of 20 G-banded metaphase spreads from reprogrammed cells will be performed.
Cellartis Spontaneous In Vitro Differentiation Service (Cat. #Y60305)
A 3-week spontaneous in vitro differentiation protocol will be used on a subset of reprogrammed cells. Cells will be fixed and immunolabelled for ASMA (mesoderm), FoxA2 (endoderm), and βIII-tubulin (ectoderm) to confirm trilineage potential.
Cellartis STR Analysis Service (Cat. #Y60306)
Genomic DNA will be isolated from reprogrammed iPS cells and the corresponding starting material. Short tandem repeat (STR) analysis will be performed on 16 loci to verify there are no differences between the samples.
Project initiation
You may wish to submit your own fibroblast or PBMC samples for Cellartis reprogramming services or use Cellartis sourcing services to find custom starting materials. If you would like us to start with your cells, we will need:
Fibroblasts: 5 vials (1 million cells per vial) of customer-provided fibroblasts, with relevant information regarding culturing and freezing protocols
PBMCs: 5 vials (10 million cells per vial) of customer-provided PBMCs, with relevant information regarding culturing and freezing protocols
For safety and QC purposes, we can only work with human samples that are free of human pathogens and that have passed a sterility test. We require documentation proving that all samples shipped to us are sterile and pathogen-free. In the absence of documentation, testing for pathogens (Cat. #Y60301) and sterility (Cat. #Y60302) will be performed via a third-party service prior to the initiation of Cellartis services. Please note that safety and sterility testing will incur additional lead time and costs.

Note: Takara Bio Europe, AB (TBEAB) will perform the reprogramming service only one time per purchase. Each purchase of the reprogramming service includes one transduction attempt with Sendai virus per donor sample. If a successful reprogramming cannot be achieved after the transduction of the donor sample with Sendai virus, the reprogramming service will be terminated and neither TBUSA nor TBEAB will have any further obligation, including additional attempts of the reprogramming service. If additional work is required, TBUSA will discuss such work, pricing, and lead time with the customer.
