With the Cellartis directed differentiation to DE cells service, we deliver high-quality, custom DE cells derived from your human ES or iPS cells. As an alternative to providing your own cells, you may request Cellartis sourcing or reprogramming services.
Our differentiation protocols start with thawing and adapting your pluripotent cells to the Cellartis DEF-CS 500 Culture System, which maintains cells in a highly pluripotent and homogeneous state. After adaptation, cells are expanded to generate a Seed Bank. The pluripotent cells are then differentiated into definitive endoderm cells, banked, and characterized.

QC testing
Included with definitive endoderm directed differentiation services:
- Visual inspection of DE cell morphology
- Viability (>6 million viable cells per vial)
- Immunocytochemistry to verify the definitive endoderm phenotype: Sox-17 (+), Oct-4 (-)
- Sterility testing (including Mycoplasma)
Project initiation
Successful differentiation depends on the quality of the starting material; a homogeneous, undifferentiated stem cell population is ideal. To get started, we will need 5 vials (2.5 million cells per vial) of customer-provided hiPS or hES cells, with relevant information, including name of the donor cell line, culture conditions, and cell density upon freezing. Your cells will be adapted to the DEF-CS system, and then the differentiation protocol can begin.
For safety and QC purposes, we can only work with human samples that are free of human pathogens and that have passed a sterility test. We require documentation proving that all samples shipped to us are sterile and pathogen-free. In the absence of documentation, testing for pathogens (Cat. # Y60301) and sterility (Cat. # Y60302) will be performed via a third-party service prior to the initiation of Cellartis services. Please note that safety and sterility testing will incur additional lead time and costs.

Note: Takara Bio Europe, AB (TBEAB) will perform the differentiation service only one time per purchase. Each purchase of the differentiation service includes protocol optimization to determine whether or not successful differentiation of the Customer’s iPS or ES cell line can be achieved. If a successful differentiation cannot be achieved after the performance of the protocol optimization, the differentiation service will be terminated and neither TBUSA nor TBEAB will have any further obligation, including additional attempts of the differentiation service. If additional work is required, TBUSA will discuss such work, pricing, and lead time with the customer.
