Cellartis human pluripotent stem cell services

Cell banking

Our scientists have over 15 years of experience with human pluripotent stem cell culture, so we know how hard it can be to grow pluripotent cells. To greatly simplify this process, we have developed a culture system that efficiently expands iPS and ES cells in a non-colony, homogeneous, monolayer culture, eliminating the need for feeder cells and complicated culture tactics.

Overview  

With Cellartis cell banking services, we will deliver high-quality Master Cell Banks from your human ES or iPS cells. Our culture protocols utilize the Cellartis DEF-CS 500 Culture System, which maintains cells in a highly pluripotent and homogeneous state. As part of our service, cells previously cultured in other systems will be adapted to the DEF-CS culture system prior to expansion and banking.

QC testing  

Included with cell banking services:

  • Visual inspection and confirmation of the morphologies of the cells in the starting material and in the Master Cell Bank
  • Immunocytochemistry of the cells in the Master Cell Bank to verify the presence of the pluripotency markers Oct-4, SSEA-4, TRA-1-60, and TRA-1-81, and the absence of SSEA-1
  • Sterility testing of the Master Cell Bank (bacteria, including Mycoplasma)

Additional testing and characterization services for banked cells:

  • Cellartis Descriptive Karyotyping Service (Cat. #Y60304)
    • Karyotype analysis of 20 G-banded metaphase spreads from banked cells will be performed.
  • Cellartis Spontaneous In Vitro Differentiation Service (Cat. #Y60305)
    • A 3-week spontaneous in vitro differentiation protocol will be used on a subset of banked cells. Cells will be fixed and immunolabeled for ASMA (mesoderm), FoxA2 (endoderm), and βIII-tubulin (ectoderm) to confirm trilineage potential.
  • Cellartis STR Analysis Service (Cat. #Y60306)
    • Genomic DNA will be isolated from banked cells and the corresponding starting material. Short tandem repeat (STR) analysis will be performed on 16 loci to verify there are no differences between the samples.

Project initiation  

You may wish to submit your own iPS or ES cells for Cellartis banking services or use Cellartis sourcing and reprogramming services to generate the starting material. If you would like use to start with your cells, we will need 5 vials (>1 million cells per vial) of hiPS or hES cells, with relevant information, including name of donor cell line, culture conditions, and cell density upon freezing.
Your cells will first be adapted to the DEF-CS Culture System, and then the expansion can begin.

For safety and QC purposes, we can only work with human samples that are free of human pathogens and that have passed a sterility test. We require documentation proving that all samples shipped to us are sterile and pathogen-free. In the absence of documentation, testing for pathogens (Cat. #Y60301) and sterility (Cat. #Y60302) will be performed via a third-party service prior to the initiation of Cellartis services. Please note that safety and sterility testing will incur additional lead time and costs.

The use of iPSCs produced by this service (hereinafter referred to as the "custom iPSCs") is strictly limited to customer’s own internal research. Customer cannot have any right to use the custom iPSCs in humans for any purposes including but not limited to diagnostics and/or therapeutics, or otherwise clinical trials. There is no right to resell or transfer the custom iPSCs to a third party regardless of whether or not compensation is received.

The use of custom iPSCs is not allowed for the purposes of the intended use as described below: 1) Creation of a human clone through transplantation of the custom iPSCs-derived embryos into human or animal individuals, 2) Custom iPSCs transplantation into human embryos, 3) Custom iPSCs transplantation into human fetuses, and 4) Production of human embryos using the custom iPSCs-derived germ cells.

Customers wishing to use the custom iPSCs for purposes other than internal research use should contact us. No express or implied license is granted to the customer. Any use of the custom iPSCs for Commercial Purposes requires a patent license from iPS Academia Japan, Inc.
