Western blot detection of Reef Coral Fluorescent Proteins by the Anti-RCFP Polyclonal Pan Antibody. This antibody detects all of the Reef Coral Fluorescent Proteins (AmCyan, AsRed, DsRed2, DsRed-Express, HcRed, ZsGreen, and ZsYellow). It does not recognize green fluorescent protein variants derived from Aequorea species (including AcGFP1). Cell lysates were prepared from HEK 293 cell lines stably expressing the indicated protein. Lysates were analyzed by Western blot using Anti-RCFP Polyclonal Pan Antibody at a 1:1,000 dilution. Each lane contains approximately 35,000 cell equivalents. Lane 1: AcGFP1. Lane 2: Blank. Lane 3: DsRed-Express. Lane 4: AmCyan1. Lane 5: ZsGreen1. Lane 6: Control (untransfected cells). Lane 7: ZsYellow1. Lane 8: AsRed2. Lane 9: HcRed1.

The Cas9 polyclonal antibody detects Cas9 protein by immunocytochemistry (ICC). RPE cells stably expressing ZsGreen1 were transfected with 0.35 µg of a plasmid encoding wild-type Cas9 under the control of the SV40 promoter using Xfect Transfection Reagent. Cells were fixed with 4% paraformaldehyde 48 hours post-transfection and stained with Cas9 polyclonal antibody (1:150) for one hour at room temperature (RT), followed by staining with donkey anti-rabbit secondary antibody(1:100) for 30 min at RT. In the control, cells were only stained with the secondary antibody.

Western blot detection of a DmrC-tagged protein with the DmrC Polyclonal Antibody (Cat. No. 635091). HeLa cells were transfected with a plasmid engineered to express a DmrC-AcGFP1 fusion protein. Lysate was prepared from the equivalent of 3.5 x 10⁴ cells and resolved on a polyacrylamide gel, followed by Western blot analysis with the DmrC Polyclonal Antibody (1:1000 dilution). A single band was detected at the expected molecular weight (Lane 3). No band was detected in lysate from untransfected HeLa cells (Lane 1) or HEK 293 cells (Lane 2).
