Reef Coral Fluorescent Protein Vector Set

Cat. # Product Size License Quantity Details
630050 Reef Coral Fluorescent Protein Vector Set Each USD $5444.00

A set of twelve vectors encoding novel fluorescent proteins in four colors: cyan, green, red and yellow. Four constructs are prokaryotic expression vectors that also serve as convenient sources of fluorescent protein cDNAs. In each of these prokaryotic vectors, the full-length cDNA is flanked by distinct MCSs to facilitate excision from the vector and insertion into other eukaryotic or prokaryotic expression constructs. Eight of the vectors are designed for studies in mammalian systems. These vectors allow expression of a protein of interest as a C- or N-terminal fusion to any of the four fluorescent proteins. The fusion vectors can also be used as cotransfection markers; the unmodified vectors will express fluorescent protein.

Lenti-X™ DD Red Reporter System

Cat. # Product Size License Quantity Details
631753 Lenti-X™ DD Red Reporter System Each USD $1096.00

The Lenti-X DD Red Reporter System includes the Lenti-X DD-tdTomato Vector Set and Shield1. The Lenti-X DD-tdTomato Vector Set includes two lentiviral expression vectors (a reporter vector and a control vector) that produce high titers of recombinant lentivirus, which can efficiently transduce both dividing and nondividing mammalian cells. The system provides enough reagents for 16 packaging reactions. The pLVX-DD-tdTomato Reporter Vector is a promoterless vector that allows you to insert your promoter of interest upstream of the red fluorescent protein tdTomato, tagged at its N-terminus with the ProteoTuner destabilization domain (DD). In the absence of the membrane-permeant ligand Shield1, the DD causes the reporter protein to be rapidly targeted to and degraded by proteasomes. This very efficient and controllable destabilization method minimizes reporter background fluorescence from leaky promoters prior to promoter activation. To analyze promoter activity, a candidate inducer is added to the medium along with Shield1, which binds to the DD tag and thereby stabilizes the reporter protein and allows it to accumulate. As a result, only the reporter molecules expressed during promoter induction will contribute to the fluorescence signal, providing a considerably higher signal-to-noise ratio than that obtained with nondestabilized or constitutively destabilized reporter systems. Cells used to monitor uninduced promoters (e.g., the negative control) will be treated with Shield1 alone.

DD-tdTomato Reporter System

Cat. # Product Size License Quantity Details
632190 DD-tdTomato Reporter System Each USD $569.00

The DD-tdTomato Reporter System includes the pDD-tdTomato Reporter vector and Shield1. The pDD-tdTomato Reporter is a promoterless vector that allows you to insert your promoter of interest upstream of the red fluorescent protein tdTomato, tagged at its N-terminus with the ProteoTuner destabilization domain (DD). The DD causes the reporter protein to be rapidly targeted to and degraded by proteasomes. This very efficient and controllable destabilization method will minimize background fluorescence from leaky promoters prior to promoter activation.

To analyze promoter activity, a candidate inducer is added to the medium along with Shield1, which effectively stabilizes the reporter protein and allows it to accumulate. As a result, only the reporter molecules expressed during promoter induction will contribute to the fluorescence signal, providing a considerably higher signal-to-noise ratio than that obtained with non-destabilized or constitutively destabilized reporter systems.

pDsRed2-1 Vector

Cat. # Product Size License Quantity Details
632405 pDsRed2-1 Vector 20 ug USD $589.00

pDsRed2-1 encodes DsRed2, a variant of the Discosoma sp. red fluorescent protein (DsRed). DsRed2 contains six amino acid substitutions that accelerate chromophore maturation and decrease protein aggregation. In addition, the coding sequence for DsRed2 contains a series of silent base-pair changes for higher expression in mammalian cells. pDsRed2-1 can be used as a reporter to monitor the activity of promoters and promoter/enhancer combinations cloned into the multiple cloning site (MCS) upstream of the promoterless DsRed2 gene. Without the addition of a functional promoter, this vector will not express DsRed2.

pAcGFP1-1 Vector

Cat. # Product Size License Quantity Details
632497 pAcGFP1-1 Vector 20 ug USD $589.00

pAcGFP1-1 encodes a green fluorescent protein (AcGFP1) from Aequorea coerulescens. The fluorescent protein coding sequence in this construct has been human-codon-optimized for efficient expression and enhanced brightness in mammalian cells. The vector contains a neomycin resistance gene for the selection of stable transfectants. pAcGFP1-1 can be used as a reporter to monitor the activity of promoters and promoter/enhancer combinations cloned into the multiple cloning site (MCS) upstream of the promoterless AcGFP1 gene. Without the addition of a functional promoter, this vector will not express AcGFP1.

pDsRed-Express-1 Vector

Cat. # Product Size License Quantity Details
632413 pDsRed-Express-1 Vector 20 ug USD $589.00

pDsRed-Express-1 is a promoterless mammalian expression vector that can be used to monitor transcription from different promoters and promoter/enhancer combinations inserted into MCS. It encodes DsRed-Express, a variant of Discosoma sp. red fluorescent protein (DsRed). DsRed-Express contains nine amino acid substitutions which improve the solubility of the protein, reduce the time from transfection to detection of red fluorescence, and decrease the level of residual green emission. When DsRed-Express is expressed in mammalian cell cultures, red-emitting cells can be detected by either fluorescence microscopy or flow cytometry 8–12 hours after transfection. Although DsRed-Express most likely forms the same tetrameric structure as wild-type DsRed, DsRed-Express displays a reduced tendency to aggregate. The DsRed-Express coding sequence is human codon-optimized for high expression in mammalian cells.
