A set of twelve vectors encoding novel fluorescent proteins in four colors: cyan, green, red and yellow. Four constructs are prokaryotic expression vectors that also serve as convenient sources of fluorescent protein cDNAs. In each of these prokaryotic vectors, the full-length cDNA is flanked by distinct MCSs to facilitate excision from the vector and insertion into other eukaryotic or prokaryotic expression constructs. Eight of the vectors are designed for studies in mammalian systems. These vectors allow expression of a protein of interest as a C- or N-terminal fusion to any of the four fluorescent proteins. The fusion vectors can also be used as cotransfection markers; the unmodified vectors will express fluorescent protein.

pTRE-Cycle2 is a bidirectional, Tet-inducible expression vector that allows you to reversibly regulate (i.e., cycle) the amount of a protein of interest in mammalian cells while coexpressing the red fluorescent protein mCherry. Expression of both proteins is tightly regulated by the bidirectional, TRE-based promoter, PTight-Bi. Expression of the gene of interest is controlled by an additional mechanism, since the resulting protein is fused to an N-terminal ProteoTuner destabilization domain (DD), which causes rapid proteasomal degradation of any protein to which it is fused. The degradation process can be reversed repeatedly by adding the stabilizing ligand Shield1 and then removing it from the culture medium. mCherry is coexpressed with the tagged protein, but does not contain a DD-tag, and thus is subject only to Tet-based regulation. The vector is intended for use with any Tet-On or Tet-Off Advanced Expression System. To select stable cell lines, the vector must be cotransfected with one of the provided linear selection markers.

This lentiviral expression vector encodes a DsRed-Express2 fluorescent protein tag. The protein is a rapidly maturing tetrameric fluorescent protein from Discosoma sp. that has minimal cytotoxicity, high solubility, and exhibits strong and stable expression in mammalian cells. Inserting a cDNA in the MCS downstream of the DsRed-Express2 coding sequence joins your protein of interest to the C-terminus of the tag, and allows the fusion protein to be tracked and studied in transduced cells. To package the vector into high-titer, replication-incompetent lentivirus, we recommend using the Lenti-X Packaging Single Shots (VSV-G) and the Lenti-X 293T Cell Line. The resulting lentivirus can then be used to transduce virtually any mammalian cell type.
