Yeast two-hybrid cDNA libraries—Mate & Plate

We strongly recommend the use of our Mate & Plate and normalized Mate and Plate Libraries with the Matchmaker Gold Yeast Two-Hybrid System. These libraries provide, by far, the simplest method for yeast two-hybrid screening because they do not require library-scale transformations or labor-intensive library amplification. Thus, very little optimization or hands-on time is required with these libraries.

We strongly recommend the use of our Mate & Plate and normalized Mate and Plate Libraries with the Matchmaker Gold Yeast Two-Hybrid System. These libraries provide, by far, the simplest method for yeast two-hybrid screening because they do not require library-scale transformations or labor-intensive library amplification. Thus, very little optimization or hands-on time is required with these libraries.

To screen, simply take a 1-ml library vial and culture overnight with a Y2H Gold clone that expresses your bait protein, and then plate on selective media. A single Mate & Plate Library aliquot is sufficient for the complete screening of more than one million independent clones.

Or do it yourself!

Alternatively you can use our Make Your Own “Mate & Plate” Library System (Cat. # 630490) to make your own customized library. With this system, you can generate enough 1 ml-library aliquots for more than 100 screens.

Interacting bait and prey fusion proteins drive the expression of four different reporters from three different GAL4-responsive promoters (M1, G1, and G2), which are stably integrated in the genome of the reporter strain, Y2H Gold. Aureobasidin A (AbA) resistance and the two auxotrophic reporters for histidine and adenine biosynthesis confer growth selection in the presence of AbA and on histidine- and adenine-deficient media, while the a-galactosidase reporter produces blue colonies in the presence of X-alpha-Gal.
