Pfu Protease S is an endo-type serine protease with broad specificity for native and denatured proteins. Cleavage occurs mainly on the carboxy terminus of peptide bonds on hydrophobic amino acid residues.

Optimum temperature: 85-95°C
Optimum pH: 6.0-8.0
Tolerance to denaturants at 95°C pH 7.0:
- 1% SDS; retains 50% activity after 24 hr at 95°C, pH 7.0
- 6.4 M urea; retains 70% activity after 1 hr at 95°C, pH 7.0
- 50% acetonitrile; retains 90% activity after 1 hr at 95°C, pH 7.0
Definition of activity
One unit of activity corresponds to the amount of enzyme required to hydrolyze 1 mmol of N-succinyl Ala-Ala-Pro-Phe p-nitroanilide in 1 minute at 95°C and pH 7.0.
