16S DNA Quantitative Standard for Microbiome
The 16S DNA Quantitative Standard for Microbiome is an artificial nucleic acid reference material designed for quantitative 16S rRNA microbiota analysis using next-generation sequencing (NGS). It is a cocktail of 12 different artificial 16S rRNA gene (rDNA) sequences, each with a predetermined copy number. Because these sequences are artificial and not found in nature, the standard can be spiked into any biological sample without interfering with the microbiota analysis.

By creating a calibration curve from the known copy numbers of the standard sequences and the reads obtained during NGS, researchers can move beyond relative abundance to the absolute quantification of bacteria in their samples. This product is also ideal for evaluating the performance of different experimental designs and for accuracy control in each test.
Overview
- Allows for the determination of the absolute copy number of bacteria in a sample, providing more insightful data than traditional relative abundance measurements
- Uses artificial 16S rRNA gene sequences that do not exist in nature, ensuring that the spike-in standard does not affect the analysis of the sample's natural microbiome
- Can be spiked into any biological sample and is compatible with NGS analysis of various variable regions of the 16S rRNA gene (including V1-V2, V3-V4, and V4), demonstrating high linearity in calibration curves across these regions
- Serves as a reliable control to evaluate the performance of experimental designs and to ensure the accuracy and consistency of each test
- Demonstrates a high correlation between samples with and without the spike-in, indicating no significant impact on the detected species
- Results in quantitative data showing a strong correlation with standards from other methods like digital PCR
