For selecting colonies, the appropriate dose must be determined empirically for each specific cell line. We recommend testing a dosage range using dishes of untransfected cells and choosing the dose that kills all of the cells in 3–5 days. If all of the cells die in less than 24 hours, you should use a lower dose.
Overview
- The most widely used selection agents for transfected mammalian cells carrying the resistance genes NeoR, HygR, and PuroR/Pac respectively
- G418 sulfate: An aminoglycoside antibiotic similar to gentamycin. Toxic to bacteria, yeast, higher plants, and mammalian cells, in addition to protozoans and helminths
- Hygromycin B: An aminoglycoside antibiotic that inhibits protein synthesis in bacteria, fungi, and higher eukaryotes
- Puromycin dihydrochloride: A broad spectrum antibiotic that inhibits protein synthesis in both prokaryotic and eukaryotic organisms
