A scalable, instrument-free approach to true single-cell spatial transcriptomics
We present a truly instrument-free spatial transcriptomics workflow that combines Trekker technology (a barcoded oligonucleotide tagging approach based on the Slide-tags method) with the Illumina single-cell 3’ RNA Prep assay to deliver high-density, high-sensitivity spatial data with true single-cell resolution.
- Single-cell boundaries in densely packed tissue are easily resolved without the need for deconvolution or cell segmentation.
- Robust detection of low-abundance and spatially restricted transcripts in complex human tissues is enabled with Trekker technology. Clinically relevant genes that are frequently diluted in spot-based methods are clearly resolved in human breast cancer at true single-cell resolution.
- Neighborhood-based niche analysis and identification of distinct multicellular microenvironments found in human breast cancer tissue is possible with single-cell spatial resolution.
